Effects on Lipid Peroxidation and Antioxidative Enzymes of Euonymus alatus in Cultured Rat Hepatocytes

Abstract: Abstract:The Euonymus alatus (Thunb.) Sieb. has long been used as a crude drug. In this paper, we investigate the effects of E. alatus on cultured hepatocyte cell system and lipid peroxidation in hydrogen peroxide (H 2 O 2 ) treatment conditions. The study covers the physiological activity (the antioxidative activity and the nitrite-scavenging effect) of E. alatus . H 2 O 2 that can produce intracellular free radical was used for inducer of the peroxidation of cellular lipids. Treatment of E. alatus attenuated… Show more

“…Similar findings were demonstrated with pheochromocytoma cell lines (PC12) [48], HUVEC cells [49], rat hepatocytes [50], bone marrow stromal cells [37], and MC3T3-E1 cells [51]. The antioxidant enzyme was used up to eliminate the lipid peroxidation products or deactivated and glycated by the radicals [52, 53].…”

“…The cytotoxic action of STZ is allied with the generation of reactive oxygen species causing oxidative damage (Szkudelski, 2001). Antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and reduced glutathione (GSH) provides support in scavenging the free radicals by breaking down the ROS (Kim JK et al, 2009). Past investigation have reported a flaw in the scavenging machinery in diabetic patients as compared to the healthy control (Anwer et al, 2012;Anwer et al, 2007).…”

“…Unfortunately, due to the complexity of organ and tissue organization in in vivo experiments, along with sudden hormonal changes and other factors, it is difficult to prove the molecular mechanism of a specific regulation. One of the most commonly used external signals increasing intracellular oxidative stress for an investigation like this one in cultured cells is H 2 O 2 (Alia et al 2005;Fukui et al 2010;Kim et al 2008;O'Brien et al 2000). H 2 O 2 damages DNA, lipids, and other macromolecules in the cell, and may lead to oxidative damage (Aherne and O'Brien 1999).…”
