Depression is partially caused by inflammation in central nervous system. This study investigated the ameliorative effects of phenol glycosides (PG) from Ligustrum lucidum Ait. (Oleaceae) on neuroinflammation and depressive-like behavior in mice hypothalamus as well as the molecular mechanism. Mice were administered with PG extract for 2 weeks prior to treatment with LPS. The mice treated with PG extract showed resistance to LPS-induced reduction in body weight and LPS-induced depressive-like behaviors shown by sucrose preference, tail suspension test, forced swimming test and open field test. LPS-induced activation of microglial cells and elevation in protein expression of inflammatory cytokines including IL-1β, RANTES and MCP-1 in hypothalamus of mice were abrogated by pre-treatment with PG extract. This extract down-regulated expression of TLR4, MyD88, NLRP3, renin and angiotensin II and decreased proportional area of Iba-1 + microglias in hypothalamus. Pretreatment with PG extract inhibited LPS-triggered activation of CaSR/G α11 signaling, stimulated 1-OHase expression in hypothalamus, and enhanced circulating 1,25 (OH) 2 D 3 level. Overall, pre-treatment with PG extract ameliorated LPS-induced depressive-like behaviors by repressing neuroinflammation in mice hypothalamus which was attributed to its suppression on activation of microglia and production of inflammatory cytokines via acting on TLR4 pathway, CaSR and RAS cascade associated with improving vitamin D metabolism. The current prevalence in US is estimated to 10-14 million annually which is more than 5% of the population (Bab & Yirmiya, 2010b). Lifetime and 12-month prevalence of major depressive disorder in China were 6.8 and 3.6%, respectively (Huang, Wang, et al., 2019). Depression has been even implicated as a possible risk factor for other chronic diseases, such as osteopenia characterized by loss of bone minerals (Schweiger et al., 2016). However limited therapeutic options for the treatment of depression are available, creating a great individual and societal burden (Li et al., 2019). Thus, development of novel antidepressant drugs is a pressing task (Zhang & Cheng, 2019).

Influence of Vitamin D Supplementation on Mental Health in Diabetic Patients: A Systematic Review

Diabetes is associated with a number of mental health consequences, including enhanced risk of depression and anxiety, as well as decreased quality of life, and vitamin D deficiency is considered to be one of the factors that influence these outcomes in diabetic patients. The aim of the present study was to conduct a systematic review of the literature presenting the data regarding the influence of vitamin D supplementation on mental health in diabetic adults. This study was conducted in accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines and registered in the International Prospective Register of Systematic Reviews (PROSPERO) database (Registration number CRD42020155779). A systematic search of the PubMed and Web of Science databases was performed, and the intervention studies published until September 2021 were included in the review. The human studies were included if an adult sample of diabetic individuals received vitamin D supplementation during the intervention and its effect on any mental health aspect was assessed, but studies presenting the influence of combined supplementation of multiple nutrients were excluded. After removing duplicate records, a total of 8,514 publications were screened and assessed independently by two researchers, based on their title, abstract, and full text. Finally, six studies were included in the current systematic review, and the risk of bias was evaluated using the Newcastle–Ottawa Scale (NOS). The included studies analyzed the influence of a specific dose of vitamin D, or different doses of vitamin D, or compared the results of supplementation with a specific dose of vitamin D against the placebo group. The supplementation was performed for at least 12 weeks. The mental health outcomes analyzed in these studies included health-related quality of life, depression, anxiety, stress, and general mental health status of adult diabetic patients. The results of the majority of the studies confirmed the positive influence of vitamin D supplementation on the mental health of diabetic individuals. Those studies that analyzed the influence of vitamin D supplementation on depression and anxiety established the beneficial effect of the vitamin. In some studies, the influence of vitamin D supplementation on the health-related quality of life was not considered unless combined with mindfulness training. However, it must be emphasized that different dosage regimens and intervention periods were followed in the reviewed studies, and only a small number of studies were randomized against placebo, which should be considered as a limitation of the present study. The findings of the conducted systematic review demonstrated the positive influence of vitamin D supplementation on the mental health of diabetic patients, which was proved for anxiety and depression, but in the case of health-related quality of life, the positive effect was observed only when the intervention included mindfulness training. These outcomes suggest that supplementation should be recommended to improve the vitamin D status and the mental health of patients in this group.

Objetivo: realizar uma revisão de literatura acerca da influência da deficiência de vitamina D, de metilfolato e de magnésio no desenvolvimento da depressão. Metodologia: Desenvolveu-se uma revisão do tipo integrativa, com estudos publicados entre 2016 e 2020. Inicialmente, estabeleceu-se a questão norteadora, seguida da seleção dos descritores “Deficiência de Vitamina D”; “ácido fólico”, “deficiência de magnésio” e “depressão”, conforme a plataforma dos Descritores em Ciências da Saúde. As bases de dados destinadas para a busca das publicações, foram: Scielo, Medline, Lilacs e Pubmed. Em seguida definiu-se os critérios exclusão e inclusão das publicações. Resultados e Discussão: Assim, a partir do protocolo metodológico empregado, 24 artigos foram selecionados para compor o estudo. A maior parte (n=19, 79,17%) correspondem a publicações que avaliaram de alguma forma a relação da vitamina D com a depressão. Quatro (16,66%) estudos correspondem à relação do ácido fólico, e um (4,17%) ao papel do magnésio nos distúrbios depressivos. Muitos autores observaram que a deficiência de vitamina D encontra-se associada à presença de sintomas depressivos. Contudo, verificou-se uma escassez de estudos científicos que visem avaliar a influência da deficiência o ácido fólico e magnésio no desenvolvimento da depressão. Conclusão: constatou-se que a maior parte estudos concentra-se na associação entre a ausência da vitamina D e depressão, demonstrando que muitos pesquisadores encaram esse micronutriente como um elemento envolvido em tal distúrbio. Entretanto, ressalta-se a necessidade de mais investigações que visem colaborar com o entendimento do papel do folato e do magnésio nas alterações de humor.
