“…Dothiepin, a TCA and a thio-analogue of amitriptyline (Rydzynski 1966), has been shown to have potent antidepressant and anxiolytic properties with minimal side effects (Lipsedge et al 1971;Johnson et al 1973;Lambourn and Rees 1974). Its antidepressant activity is mediated through facilitation of noradrenergic neurotransmission by uptake inhibition (Ishikawa et al 1986) and possibly also by enhancement of serotoninergic neurotransmission (Lancaster and Gonzalez 1989). The anticholinergic actions of TCAs such as dry mouth, constipation, dizziness, tachycardia and palpitation are less pronounced with dothiepin than imipramine and amitriptyline (Lambourn and Rees 1974;Sim et al 1975).…”
