Effects of Transient Hypoxia versus Prolonged Hypoxia on Satellite Cell Proliferation and Differentiation In Vivo

Abstract: The microenvironment of the injury site can have profound effects on wound healing. Muscle injury results in ischemia leading to short-term local hypoxia, but there are conflicting reports on the role of hypoxia on the myogenic program in vivo and in vitro. In our rat model of mitochondrial restoration (MR), temporary upregulation of mitochondrial activity by a cocktail of organelle-encoded RNAs results in satellite cell proliferation and initiation of myogenesis. We now report that MR leads to a transient hyp… Show more

“…HIF-1a plays an important role in maintaining the quiescence of Pax7 + satellite stem cells and preventing their proliferation and differentiation through inhibition of the WNT pathway (42)(43)(44). To test whether HIF-1a affects the expression levels of WNT target genes in MRL/MpJ mouse MDSPCs, we measured the expression levels of these genes in DMOG-treated MRL/MpJ MDSPCs or cells cultured in hypoxia for 24 h (Fig.…”

“…A recent study proposed another approach to induce either transient or prolonged hypoxia during skeletal muscle regeneration in rats (47). Six days after injury of the quadriceps muscle caused by a needle, a cocktail of mitochondrion‐targeted RNAs was injected at the injury site.…”

“…Additional experiments are needed to determine to which extent the injured fibers are exposed to low O 2 levels in this model under normoxic conditions. A recent study proposed another approach to induce either transient or prolonged hypoxia during skeletal muscle regeneration in rats (47). Six days after injury of the quadriceps muscle caused by a needle, a cocktail of mitochondrion-targeted RNAs was injected at the injury site.…”

“…Additionally, analysis of the oxygen uptake of stem cells revealed shifts in their energy metabolism. These shifts were correlated with the phenotypic changes observed in the study of inherited diseases , wound healing , and differentiation pathways . Therefore, the capacity to monitor the DO in a culture and to quantify the oxygen kinetics of the cells, such as the specific oxygen uptake rates (sOUR), is critical to the development of stem cell‐based therapies.…”
