Effects of Topical Calcipotriol on Calcium Metabolism in Psoriatic Patients: Comparison with Oral Calcitriol

Abstract: The analogue of calcitriol calcipotriol (MC 903 Daivonex®) has been proven effective in the treatment of psoriasis, when given topically. However, the possible influence of cutaneously absorbed MC 903 on calcium metabolism is still unclear. We evaluated various parameters of calcium metabolism in 17 psoriatic patients treated for 5.4 ± 2.3 (mean ± SD) weeks with MC 903, on 16 ± 6% of the body surface. The dose administered (100 g of Daivonex corresponding to 5 mg of MC 903) decreased the PASI score by 40.9 ± 2… Show more

“…Mean values were within normal range after calcipotriol therapy, particularly 24-hour cal cium excretion which is a reflection of intestinal absorption and the fasting calcium-to-creatinine ratio which evaluates bone resorption. These parameters were not, for cost reasons, systemati cally evaluated in patients of groups 2 and 3, since this had been done previously in a similar type of patients [5].…”

“…In a previous study, we found that topical calcipotriol did not modify calcium metabolism when used in patients with psoriasis covering 16±6% of the body surface and receiving 25 g of ointment per week [5]. In contrast, 1.5 jig/ day of calcitriol orally for 7 days in the same patients in duced a significant increase in plasma levels of proteinadjusted calcium and in 24-hour urinary calcium excretion, as well as a significant fall in PTH [5|.…”

“…In the study by Rico et al (8), 10 patients with psoriasis presented raised serum PTH levels during treatment with oral calcitriol. Other studies have failed to detect an alteration in PTH serum levels in the course of different treatments (9)(10)(11). In the present study, we measured the serum PTH levels in a larger group of patients with psoriasis and in comparison to a control group.…”
