Effects of the extracellular matrix on lumican expression in rat aortic smooth muscle cells in vitro

Abstract: Lumican is a small leucine-rich proteoglycan (SLRP), which contributes to cell migration, proliferation, tissue hydration, and collagen fibrillogenesis. Whether lumican is localized in rat aortic smooth muscle cells (SMCs) and what its relationships might be to other extracellular matrix components have not yet been elucidated. In this study, using reverse transcription-polymerase chain reaction (RT-PCR), competitive RT-PCR, and western blot, lumican messenger ribonucleic acid (mRNA) was expressed in cultured … Show more

“…While endoglin is poorly expressed, lumican is highly expressed in the endothelium layer of a renal vein. In this tissue, staining of lumican in smooth muscle cells could also be detected, in agreement with previous reports (Chakravarti et al, 1998;Qin et al, 2001). These results provide evidence for an inverse correlation in the tissue distribution of endoglin and lumican in vivo.…”

“…Immunoblotting for lumican (Fig. 5C) revealed multiple diffuse bands predominantly from 50 -100 kDa, indicative of a proteoglycan form of lumican (33)(34)(35)(36). Consistent with the analysis of the MS signal intensities, there was significantly (p Ͻ 0.01) more lumican present in the intima of the atherosclerosis-prone internal carotid artery than in the atherosclerosis-resistant internal thoracic artery, with the mean value elevated by ϳ3-fold.…”

“…Northern Blot and in Situ Hybridization-Total RNA from the embryonic, larval, pupal, or adult stage of Drosophila was extracted using the TRIzol reagent (Invitrogen) (37). A total of 12.5 g of RNA was separated by electrophoresis on a formaldehyde-agarose gel and transferred overnight to a Hybond-N membrane (Amersham Biosciences).…”
