Effects of the Anti-Platelet Aggregation Drug Dilazep on Cognitive Function in Dahl Salt-Sensitive Rats.

Abstract: Among the consequences of the increasing prolongation of lifespan is a worldwide increase in the number of cases of dementia or impaired cognition. In the present study, to test the hypothesis that mechanisms independent of high blood pressure are involved in maintaining cognitive function, we assessed the effects of long-term dilazep treatment on cognitive dysfunction in normotensive Dahl salt-sensitive (Dahl S) rats fed a low-salt diet, using the standard passive avoidance test. Normotensive Dahl S rats fed … Show more

“…Recently, Yamagishi et al [22] reported that platelet activation plays a major role in the development and progression of rat diabetic nephropathy. Numabe et al [23] reported that treatment with dilazep through vascular and nonvascular effects maintains brain function in animals susceptible to vascular injury and organ dysfunction. Dilazep dihydrochloride is commonly used in patients with IgA nephropathy, ischaemic heart disease, polycystic kidney disease or diabetic nephropathy [8,21,24,25], and it is known to cause an increase in cerebral blood flow [6].…”

“…Therefore, some platelet-derived miRNAs may play a potential role in platelet-associated pathologic processes, including vascular lesions. In a similar context, recent studies have reported enhanced platelet activation and aggregation in MCI in correlation with AD progression, and an ameliorating effect of anti-platelet drugs on cognitive decline [44,45]. Based on these results, we hypothesized that some miRNAs in platelets are important in the process of pathologic changes in Aβ(+)MCI patients that are known to progress to AD faster than Aβ(−)MCI patients.…”
