Effects of Temperature, pH and Blood Storage on Osmotic Fragility of Duck Erythrocytes

Abstract: Summary The osmotic fragility of duck erythrocytes at different pH and temperatures and during blood storage was studied. The fragility increased in acidic solution and also as the temperature increased. However, storage of blood decreased the fragility of duck erythrocytes.

“…A maximum of two freeze / thawing cycles was allowed since three or more cycles modify the oxidative status of the blood [20] and storage increases the levels of the markers of oxidative damage [21]. The plasma was assayed for superoxide dismutase activity, catalase activity and malondialdehyde (MDA) level.…”

“…Haematological parameters in birds have been shown to be influenced by various factors such as age, sex, season and nutrition. Packed cell volume (PCV), haemoglobin (Hb) concentration and red blood cell (RBC) count have been reported to increase with age in chickens (Islam et al, 2004), Pigeons (Columba livia forma domestica) (Pavlak et al, 2005), Turkeys (Oyewale &Ajibade, 1990) and Budgerigars (Melopsittacus undulatus) (Harper & Lowe, 1998). Packed cell volume (PCV) and Hb values have also been reported to be higher in males than in females in Turkeys (Oyewale & Ajibade, 1990) and Pigeons (Pavlak et al, 2005).…”

Similarly, seasonal fluctuations in PCV and Hb concentrations have been reported in Pigeons in the tropics (Oladele et al, 2001b) and in the cold temperate region (Pavlak et al, 2005).…”

“…The osmotic fragility test is most frequently used to evaluate the response of erythrocytes to osmotic pressure [12] and determines their resistance to hypotonicity and hemolysis [13]. Osmotic fragility is affected by extrinsic factors such as pH, temperature, osmolality and blood storage [14][15][16][17] and intrinsic factors such as (i) presence of osmotically active constituents (ii) modifications of the lipids and proteins in the membrane [18]. The osmotic fragility provides a guide to assess the membrane integrity [19,20] and can echo the changes in membrane properties in the erythrocytes [21].…”
