Effects of tachyplesin and n-sodium butyrate on proliferation and gene expression of human gastric adenocarcinoma cell line BGC-823

Abstract: Tachyplesin as a differentiation inducer of tumor cells has similar effects as n-sodium butyrate on proliferation of tumor cells, expression of correlative oncogene and tumor suppressor gene. It also has a synergistic effect on differentiation of tumor cells.

“…Some reports have also shown alterations in gene expression of cancer cells in cell death process. For instance, tachyplesin decreased the expression of oncogenes C‐MYC, C‐ERB, and MTp53 in a human gastric adenocarcinoma cell line, and increased gene expression of p16 a tumor suppressor [Li et al, ; Shi et al, ]. In another study, lipopolysaccharide ‐ induced the expression of >40 genes in macrophage; the cationic AMP cecropin‐melittin selectively blocked lipopolysaccharide‐induced gene expression in murine macrophage cell line [Scott et al, ].…”

“…Previous studies in Caco-2 cells and lymphoma cell lines (JeKo-1, Hbl-2 and Grant-519) have reported arrests in both G 0 -G 1 and G 2 -M phases following butyrate treatment. [30][31][32][33] However, many have reported arrests in G 0 -G 1 10,14,34 or G 2 -M alone 33,35 in other cell types, such as breast and gastric cancer cells. Therefore, the timing of cell cycle arrest appears dependent on the cell line used.…”

“…A synthetic peptide from tachyplesin I has been demonstrated to decrease the growth of tumor cells in vitro and in vivo following linkage to the integrin homing arginine-glycine-aspartic acid (RGD) domain (3). Several studies had demonstrated that tachylesin inhibited the proliferation of tumor cells, including gastric adenocarcinoma, human hepatocarcinoma, prostate cancer and melanoma (3)(4)(5). In addition, tachyplesin I affects the differentiation of tumor cells (6).…”
