Stress factors are so numerous and so diverse in their strength and duration that the consequences on animal welfare can be quite varied. The first important distinction concerns the characterization of acute and chronic stress conditions. Acute stress is a short-lived negative situation that allows a quick and quite complete recovery of the physiological balance (adaptation), while chronic stress is a long lasting condition from which the subject cannot fully recover (maladaptation). In the latter case, the direct effects of the stress factors (heat, low energy, anxiety, suffering etc.), as well as the indirect ones (changes occurring at endocrinological, immune system or function level) can be responsible for pre-pathological or pathological consequences which reduce animal welfare. To evaluate the possible chronic stress conditions in single animals or on a farm (in particular a farm of dairy cows), some parameters of the direct or indirect effects can be utilised. They are physiological (mainly hormone changes: cortisol, β-endorphin), behavioural (depression), biochemical (metabolites, acute phase proteins, glycated proteins etc.), as well as performance parameters (growing rate, milk yield, fertility, etc.). Special attention has been paid to the interpretation of cortisol levels and to its changes after an ACTH challenge. Despite fervent efforts, well established and accepted indices of chronic stress (distress) are currently lacking; but without this objective evaluation, the assessment of animal welfare and, therefore, the optimization of the livestock production, could prove more difficult.

ABSTRACT. Spermatogenesis, is a complex process of precisely regulated intracellular events, where it is affected by many factors. Long-distance transport of animals is one of the stressors that may influence spermatogenesis and sperm quality. The present study chose luteinizing hormone receptor (LHR), androgen receptor (AR), and heatshock protein 70 (HSP70) as our target genes to investigate their mRNA and protein expression in the testes of long-distance transported (about 1000 km) mice. Histological analysis showed that there was a reduction in the thickness of the seminiferous epithelium in the transported mice, and a significant decrease in body weight and sperm count in the epididymis was also observed. mRNA expression was determined by QPCR in the testis of transported and control mice. The levels for AR decreased significantly in transported mice. LHR and HSP70 expression in the testes of the transported mice was slightly higher than that of control mice but did not reach a significant level. A similar tendency of protein expression was also observed by Western blot analysis. The levels of LHR and HSP70 increased slightly after transportation. However, none of the changes were statistically significant compared with the control mice. In conclusion, long-distance transport has an adverse effect on reproductive organs and spermatozoa in adult mice.

In order to study the possibility of using lymphocyte subsets as a stress indicator, the changes of the subset proportion in peripheral blood were examined after transportation. Six 3-month-old male Holstein calves were transported for about 3 h in a lorry via an ordinary road. Blood samples were collected at the expected unloading time on the day before transport (T0), immediately after unloading (T1), 24 h after unloading (T2), and 1 week after unloading (T3). CD2, CD4, CD8, CD21, CD25, WC1, and NKp46 antigens on peripheral blood mononuclear cells were measured by flow cytometry. There was an increase in the proportions of CD25+ cells and WC1+ γδT cells at T1 and T2. The proportions of NK cells increased at T1, including an increase in the population of CD2-NK cells. CD2+ NK cells did not show any change. The proportions at T1 were lower than at T3 in CD21+ B cells. CD4+ and CD8+ αβT cells did not change significantly, although CD2+ T cells showed a trend toward reduction at T1. Consequently, CD2-NK cells and WC1+ γδT cells might offer potential as stress index for calf transport.
