Effects of renal denervation of the contralateral kidney on blood pressure and sodium and eicosanoid excretion in the chronic phase of two-kidney, one-clip renovascular hypertension in rats

Abstract: Afferences from the contralateral kidney appear to participate in the maintenance of 2K-1C renovascular hypertension due to the activation of central mechanisms regulating blood pressure.

“…Other rodent models of CKD include one‐kidney one‐clip, two‐kidney one‐clip, streptozotocin‐induced diabetes, unilateral ureteral obstruction, genetic models of diabetes and diabetic nephropathy such as the diabetic obese db/db mouse, the obese ob/ob mouse, the hypoinsulinaemic non‐obese diabetic mouse, the KKAy mouse and the New Zealand obese mouse . Two‐kidney one‐clip and one‐kidney one‐clip models also require surgery, with the one‐kidney one‐clip model requiring unilateral nephrectomy.…”

“…Increase in blood pressure can be abolished in a Goldblatt-hypertension animal model ("2 kidney 1 clip") if the affected kidney is denervated [57]. Interestingly, denervation of the non-affected contralateral kidney also abolishes hypertension in this model [58].…”

“…High blood pressure does not always normalize completely after operative correction of renal arterial stenosis or nephrectomy of the stenotic kidney in chronic two-kidney renovascular hypertension [1,2]. The mechanisms that contribute to the maintenance of hypertension are still unclear.…”

“…In individual cases, only the removal of the contralateral kidney led to a complete normalization of blood pressure [1]. Activation of central pressure mechanisms by renal afferents from the contralateral kidney is one explanation for the failure of normalization of blood pressure during the chronic phase of two-kidney, one-clip renovascular hypertension (2K-1C RVH) after correction of the arterial stenosis [1,8,9]. In addition, eicosanoids have been implicated in this phenomenon [4Á6].…”
