Effects of radiation on the expression of adhesion molecules and cytokines in a static model of human dermal microvascular endothelial cells

Abstract: BACKGROUND: Radiation-induced wound healing complications represent an important clinical problem. Microvascular compromise is an important component of its pathogenesis and the microvascular endothelial cell is the key representative affected at the cellular level. MATERIAL AND METHODS: Human dermal microvascular endothelial cells (HDMEC) were cultured and irradiated with doses of 2 to 12 Gy. Cell density was determined 48 h after radiation using a semi-automated cell counting system. Levels of interleukin-6 … Show more

“…The other explanatory model is focused on fibroblast dysfunction because fibroblasts from irradiated tissues seem to be characterized by impaired proliferation rates [ 24 ]. These theories are supported by the experimental results of our present study on fibroblast proliferation and our previous results concerning endothelial dysfunction after external radiation [ 11 ].…”

“…Previous investigations of our group were focused on effects of external radiation on HDMECs [ 11 , 15 ]. The present study revealed a significant reduction of cell proliferation after external radiation of NHFs.…”

“…The current study demonstrated that over a large exposure gradient, the initial radiation dose appears to be directly correlated to the degree of microvascular injury. This finding is consistent with results by Haubener et al who showed that varying doses of single fraction radiation exposure had an increasing effect on diminishing endothelial cell number in vitro ( 26 ).…”

“…Dies kann zu einer anhaltenden entzündlichen Gewebeveränderung mit dem Auftreten chronischer Wunden führen. Anhand von Zellkulturmodellen konnte unsere Arbeitsgruppe bereits die proinflammtorischen Zellreaktionen nach externer Bestrahlung dokumentieren [8,9]. Rigotti et al [ tischen Gesichtschirurgie [23,24] seit mehr als 20 Jahren dokumentiert und fanden bereits im 19.…”
