Effects of proflavine on papain's catalytic activity. Proflavine-mediated decrease in KM

Abstract: Proflavine is a noncompetitive activator of papain which induces a 2.7-6.0-fold decrease in Km for the hydrolysis of ¿V,a-benzoyl-L-arginine ethyl ester and /V-benzoyl glycine ethyl ester. Proflavine alters Fmax by less than 50%. Dissociation constants (Kd) for papain-proflavine complexes were determined fluorometrically by observing the quenching of proflavine fluorescence which accompanies formation of a papain-proflavine complex.
