Polyploids play an important role in the breeding of plant for superior characteristics, and many reports have focused on the effects upon photosynthesis from polyploidization in some plant species recently, yet surprisingly little of this is known for barley. In this study, homozygous diploid and tetraploid plants, derived from microspore culturing of the barley cultivar “H30,” were used to assess differences between them in their cellular, photosynthetic, and transcriptomic characteristics. Our results showed that tetraploid barley has the distinct characteristics of polyploids, namely thicker and heavier leaves, enlarged stomata size or stomatal guard cell size, and more photosynthetic pigments and improved photosynthesis (especially under high light intensity). This enhanced photosynthesis of tetraploid barley was confirmed by several photosynthetic parameters, including net photosynthetic rate (Pn), stomatal conductance (Gs), intercellular CO2 concentration (Ci), transpiration rate (Tr), maximum net photosynthetic rate (Pmax), light saturation point (LSP), maximum RuBP saturated rate carboxylation (Vcmax), and maximum rate of electron transport (Jmax). Transcriptomic analyses revealed that just ~2.3% of all detected genes exhibited differential expression patterns [i.e., differentially expressed genes (DEGs)], and that most of these – 580 of 793 DEGs in total – were upregulated in the tetraploid barley. The follow-up KEGG analysis indicated that the most enriched pathway was related to photosynthesis-antenna proteins, while the downregulation of DEGs was related mainly to the light-harvesting cholorophyII a/b-binding protein (Lhcb1) component, both validated by quantitative PCR (qPCR). Taken together, our integrated analysis of morphology, photosynthetic physiology, and transcriptome provides evidences for understanding of how polyploidization enhances the photosynthetic capacity in tetraploids of barley.

Background Inter-specific hybridizations were common and can easily take place in Buddleja , and it was an important way for evolution and rapid speciation. The F1 hybrid in this study was a newly identified inter-specific hybridization between B. crispa and B. offic inalis in Sino-Himalayan region. In the natural hybrid zones, F1 hybrids always occupy different habitats from their parents. The objective of this study was to explore environmental acclimatization of F1 hybrids and their parents at physiological and biochemical levels.Results The results showed that F1 hybrids performed as an intermediate in adaptation to their parents, with divergent gas-exchange and chlorophyll fluorescence features. F1 hybrids showed the parallel light compensation point and light saturation point with their parents, but low utilization efficiency to low-light density. They synthesized the greatest total chlorophyll content (10.41 ± 0.56 mg•g -1 ) in leaves than their parents. During the diurnal variation of photosynthesis, F1 hybrids markedly decreased and preserved the stomatal conductance and leaf transpiration rate at a low level. However, they kept high carbon assimilation rate and water-use efficiency with markedly increased vapor pressure deficit. In F1 hybrids, the maximum net photosynthetic rate, maximum water-use efficiency and maximum vapor pressure deficit were 10.48 ± 0.50 mmol CO 2 •mmol -1 photo, 21.52 ± 2.20 µmol•mmol -1 and 4.18 ± 0.55 kPa, respectively. In addition, all Buddleja species performed well and grow healthy with high level of the maximum photochemical efficiency of PSII and low non-photochemical quenching, 0.83 ± 0.004 - 0.85 ± 0.004, and 1.22 ± 0.15 - 1.97 ± 0.08, respectively. In F1 hybrids, they showed great photochemical activity compared to their parental species with high photochemical quenching. Furthermore, the effective quantum yield and electron transport rate presented a similar behavior.Conclusions The results indicated that F1 hybrids have great photochemical activities and growth acclimatization compared to their parents. Associated with the growth performance of F1 hybrids in the homogenous garden, our results suggested that the divergent gas-exchange and chlorophyll fluorescence patterns may facilitate F1 hybrids to respond to different habitats, and to improve growth performance.

Premise of studyIncreased genomic “material costs” of nitrogen (N) and phosphorus (P) atoms inherent to organisms with larger genome sizes (GS) has been proposed to limit growth under nutrient scarcities and promote growth under nutrient enrichments. Such responsiveness may reflect a nutrient‐dependent diploid versus polyploid advantage that could have vast ecological and evolutionary implications, but direct evidence that material costs increase with ploidy‐level and/or influence cytotype‐dependent growth, metabolic, and/or resource‐use tradeoffs is limited.MethodsWe grew diploid, auto‐tetraploid, and auto‐hexaploid Solidago gigantea plants under one of four ambient and enriched N:P treatments and measured traits related to material costs, primary and secondary metabolism, and resource‐use.Key resultsRelative to diploids, polyploids invested more N and P into cells and tetraploids grew more following N‐enrichments, suggesting that material costs increase with ploidy‐level. Polyploids also generally exhibited strategies that could minimize material‐cost‐constraints over both long (reduced monoploid GS) and short (more extreme transcriptome downsizing, reduced photosynthesis rates and terpene concentrations, enhanced N‐use efficiencies) evolutionary time periods. Furthermore, polyploids had lower transpiration rates but higher water‐use‐efficiencies than diploids, both of which were more pronounced under nutrient‐limiting conditions.ConclusionsCollectively we found that NP material costs increase with ploidy‐level but that material‐cost‐constraints might be lessened by organismal resource allocation/investment mechanisms that can also alter ecological dynamics and selection. Our results enhance mechanistic understanding of how global increases in nutrients might provide a release from material‐cost‐constraints in polyploids that could impact ploidy (or GS)‐specific performances, cytogeographic patterning, and multispecies community structuring.This article is protected by copyright.
