Effects of Phytoestrogen Extracts Isolated from Rye, Green and Yellow Pea Seeds on Hormone Production and Proliferation of Trophoblast Tumor Cells Jeg3

Abstract: Background: Phytoestrogens are a diverse group of non-steroidal plant compounds. Because they have chemical structures similar to estrogens they are able to bind on estrogen receptors in humans. Objectives: In this study, we tested the effects of crude phytoestrogen extracts from rye (Secale cereale), green pea (Pisum sativum) and yellow pea seeds (Pisum sativum cv.) on cell proliferation and the production of progesterone in trophoblast tumor cells of the cell line Jeg3. Methods: Isoflavone extracts from gree… Show more

“…This cell line maintains characteristics of first trimester trophoblast cells (Matsuo and Strauss 1994) and has been used to assess biological effects of exposures to environmental toxicants (Bergemann et al 2003; Canettieri et al 2008; Ding et al 2011; Koc et al 2003; Matscheski et al 2006; Adebambo et al 2015; Guan et al 2013; Kummu et al 2012; Alvarez and Chakraborty 2011). Cells were grown in Dulbecco’s Eagle Minimum Essential Media with 10% FBS, sodium pyruvate, and penicillin/streptomycin at 37°C with 5% CO 2 .…”

“…Genistein in comparison to daidzein was a more sufficient inhibitor for the production of hCG [13]. In addition, our former investigation on trophoblast tumor cells with different phytoestrogens showed effects on cell proliferation and hormone production [13][14][15][16][17].…”

“…For the determination of estradiol in the culture medium, a competitive enzyme immuno-assay (EIA) was applied as described previously [29]. The measurements were performed using an automated Immulite 2000 (DPC Biermann, Freiburg, Germany) hormone analyzer.…”
