Inhalant misuse is a worldwide problem, especially among adolescents. Toluene is the most widely misused inhalant. One hallmark of adolescence is the emergence of sexual behaviour, which can be affected by drug use. The aim of this study was to analyse the effects of toluene inhalation on different aspects of male rats' sexual behaviour using a binge pattern of exposure in adolescent rats. Male Wistar rats were individually exposed to air or 6,000 ppm toluene for 30 min (acute exposure; n = 8 each) or twice a day for 12 days (repeated exposure; n = 9 each) in static exposure chambers. Independent groups of sexually experienced, adolescent rats (postnatal day 63, PN63) were tested after acute toluene exposure for copulation, sexual incentive motivation or noncontact erections. Sexually naïve, adolescent rats (PN45-59) were repeatedly exposed to toluene and tested for sexual behaviour after completing the 1st, 3rd, 5th, 7th, 9th, 11th and 13th exposure sessions. Acute toluene exposure impaired copulatory performance, diminished sexual incentive motivation and delayed noncontact erection occurrence in sexually experienced rats. Repeated toluene exposure during adolescence completely inhibited the onset of copulatory behaviours in sexually naïve rats, at the time at which they should have appeared. However, once the inhalant exposure ended, copulatory responses appeared gradually, and animals attained a typical, stable copulatory pattern. In conclusion, acute toluene exposure impairs sexual behaviour in young, sexually experienced animals, while repeated toluene exposure during adolescence prevents the onset of copulatory behaviour, although this effect is transitory.

ObjectivesThere is limited data regarding the toxicity of methylcyclohexane, despite its wide use in rubber adhesives, paint diluents, and cleansing agents. This study aimed to verify the toxicity and influence on the reproductive system of methylcyclohexane after its repeated injection in Sprague Dawley (SD) rats.MethodsMethylcyclohexane was injected subcutaneously into male and female SD rats once a day, five times a week, for 13 weeks at different doses (0, 10, 100, and 1,000 mg/kg/day) for each group. The toxicity of testing material was verified by observing the change in body and organ weight, hematological change, pathological findings, and effect on the reproductive system at each different concentration.ResultsIn the 1,000 mg/kg/day group, there were cases of animal deaths. In animals that survived, hematological changes, including a decrease in the red blood cell count, were observed. A considerable weight gain or loss and pathological abnormalities in the liver, kidney, and other organs were found. However, the 10 and 100 mg/kg/day groups did not cause deaths or other specific abnormalities. In terms of reproductive toxicity, there were changes in hormone levels, including a significant decrease in hormones such as estradiol and progesterone (p < 0.001) in male animals. Menstrual cycle change for female animals did not show concentration dependency.ConclusionWhen injected repeatedly for 13 weeks, methylcyclohexane proved to be toxic for the liver, heart, and kidney at a high dose. The absolute toxic dose was 1,000 mg/kg/day, while the no observed adverse effect level was less than 100 mg/kg/day. The substance exerted little influence on the reproductive system.

This study evaluates the potentiality of an herbal plant Tephrosia purpurea (Linn.) Pers. in the treatment of Letrozole induced polycystic ovary syndrome (PCOS) using female Albino wistar rats. Seed powder of T. purpurea at concentration of 200 mg/kg body weight with milk as a carrier vehicle 3ml/kg body weight, given for at least 3 consecutive estrous cycles. Vaginal smear, FSH, LH, Testosterone, and Estrogen were analyzed to determine the fluctuations in sex steroid level in PCOS induced rats. The plasma testosterone and estrogen level were found to be significantly increased in rats with PCOS whereas Follicle stimulating hormone and Leutinizing hormone did not show any changes. When compared with control the PCOS induced rats showed characteristic ovary with high incidence of ovarian cyst. All the parameters assessed were significantly improved after the treatment with T. purpurea and achieved level close to 80% normalcy. Effect of T. purpurea significantly reduced histopathological changes in ovary and endocrinological and biochemical changes induced by hyperandrogenism. Further to check ovulation and fertility female rats were mated and pregnancy was confirmed. Thus potential of T. purpurea in the treatment of PCOS using an animal model suggested being a good alternative therapy in the treatment of PCOS.
