Ozone (O 3 )-induced cell death in two suspension-cultured cell lines of tobacco (Nicotiana tabacum L.) derived from Bel-W3 (hyper-sensitive to O 3 ) and Bel-B (highly tolerant to O 3 ) varieties were studied. By exposing the newly prepared cell lines to the pulse of ozonized air, we could reproduce the conditions demonstrating the difference in O 3 sensitivity as observed in their original plants, depending on the exposure time. Since O 3 -induced acute cell death was observed in the dark, the requirement for photochemical reactions could be eliminated. Addition of several ROS scavengers and chelators inhibited the cell death induced by O 3 , indicating that singlet oxygen ( 1 O 2 ), hydrogen peroxide (H 2 O 2 ), hydroxyl radical and redox-active metals such as Fe 2+ play central roles in O 3 -induced acute damages to the cells. As expected, we observed the generation of 1 O 2 and H 2 O 2 in the O 3 -treated cells using chemiluminescent probes. On the other hand, an NADPH oxidase inhibitor, superoxide dismutase (SOD), and some SOD mimics showed no inhibitory effect. Thiols added as antioxidants unexpectedly behaved as prooxidants drastically enhancing the O 3 -induced cell death. It is noteworthy that some ROS scavengers effectively rescued the cells from dying even treated after the pulse of O 3 exposure, confirming the post-ozone progress of ROS-dependent cell death mechanism. Since one of the key differences between Bel-B and Bel-W3 was suggested to be the capacity for ROS detoxification by catalase, the endogenous catalase activities were compared in vivo in two cell lines. As expected, catalase activity in Bel-B cells was ca. 7-fold greater than that in Bel-W3 cells. Interestingly, Ca 2+ chelators added prior to (not after) the pulse of O 3 effectively inhibited the induction of cell death. In addition, increases in cytosolic Ca 2+ concentration sensitive to Ca 2+ chelators, ion channel blockers, and ROS scavengers were observed in the transgenic Bel-W3 cells expressing aequorin, suggesting the action of Ca 2+ as a secondary messenger initiating the oxidative cell death. The O 3 -induced calcium response in Bel-W3 cells was much greater than Bel-B cells. Based on the results, possible pathways for O 3 -dependent generation of the lethal level of ROS and corresponding signaling mechanism for induction of cell death were discussed.

O ozônio troposférico causa necroses foliares em Nicotiana tabacum. Cultivares de tabaco são muito utilizadas como bioindicadoras de ozônio, em programas de biomonitoramento da qualidade do ar. Dentre elas destacam-se as cultivares Bel-W3 (sensível) e Bel-B (tolerante). Este estudo teve como objetivo verificar se as duas cultivares apresentam variações na estrutura foliar que possam ajudar a explicar a diferença na sensibilidade ao ozônio que ambas apresentam. Em plantas que cresceram em ambiente isento de poluentes aéreos foram coletadas folhas do terceiro nó e que foram processadas segundo técnicas usuais empregadas em anatomia vegetal. Realizaram-se análises quantitativas e qualitativas dos tecidos foliares. A cultivar Bel-W3, quando comparada à Bel-B, apresentou menor densidade estomática na superfície abaxial e maior espessura do mesofilo. Qualitativamente as células de ambas cultivares mostraram aspecto sadio, com os vacúolos ocupando praticamente todo o volume celular. A cultivar sensível apresentou visivelmente mais espaços intercelulares, câmaras estomáticas maiores e estômatos ligeiramente salientes em ambas superfícies foliares. Tais características, que facilitam a entrada e a difusão dos poluentes na folha, podem explicar, do ponto de vista estrutural, a maior sensibilidade da cultivar Bel-W3. Tropospheric ozone produces symptoms such as leaf injury in Nicotiana tabacum. Tobacco cultivars are often used as ozone indicators in air-quality biomonitoring programs. Bel-W3 (sensitive) and Bel-B (tolerant) cultivars are better known. The aim of this study was to verify if the two cultivars show variation in leaf structure that may help to explain differences in ozone sensitivity. Expanded leaves from plants growing in pollutant-free air were collected, processed according to the usual techniques of plant anatomy, and analyzed quantitatively and qualitatively. The sensitive cultivar showed lower stomatal density on the abaxial leaf surface and thicker mesophyll cells when compared to the tolerant cultivar. Qualitatively speaking, both cultivars had healthy cells with unaltered vacuoles. In the sensitive cultivar, we could see more intercellular spaces, larger substomatal chambers and slightly prominent stomata on both leaf surfaces. From a structural view point, these features explain why cultivar Bel-W3 is more sensitive.

In Arabidopsis thaliana, oxidant-induced signalling has been shown to utilize the mitogen-activated protein kinase (MAPK), AtMPK6. To identify proteins whose accumulation is altered by ozone in an AtMPK6-dependent manner we employed isotope-coded affinity tagging (ICAT) technology to investigate the impact of AtMPK6-suppression on the protein profiles in Arabidopsis both before (air control) and during continuous ozone (O 3 ) fumigation (500 nL L -1 for 8 h). Among the 150 proteins positively identified and quantified in the O 3 -treated plants, we identified thirteen proteins whose abundance was greater in the AtMPK6-suppressed genotype than in wild-type (WT). These include the antioxidant proteins, monodehydroascorbate reductase, peroxiredoxin Q, and glutathione reductase. A further eighteen proteins were identified whose abundance was lower in the ozone-treated AtMPK6-suppressed line relative to ozone-exposed WT plants. These predominantly comprised proteins involved in carbohydrate-, energy-, and amino acid metabolism, and tetrapyrrole biosynthesis. In control plants, five proteins increased, and nine proteins decreased in abundance in the AtMPK6-suppressed genotype compared to that of the WT, reflecting changes in the protein composition of plants that have AtMPK6 constitutively suppressed. Since a number of these proteins are part of the redox response pathway, and loss of AtMPK6 renders Arabidopsis more susceptible to oxidative stress, we propose that AtMPK6 plays a key role in the plant's overall ability to manage oxidative stress.
