Effects of Ovarian Steroids on Vaginal Smears in the Rat

Abstract: A correspondence between the appearance of vaginal smears and the layers of the epithelium from which the cells had desquamated was established in untreated rats during the estrous cycle, in control ovariectomized rats and in spayed rats injected with either estrogen or progesterone. The technique for preparing and staining the smears (modified Shorr’s staining procedure) is outlined. A simplified.system of classification which allows the accurate identification of the various stages of the reproductive state … Show more

“…Rats that exhibited at least 7 days of atrophic vaginal smears (Montes & Luque 1988) were injected s.c. daily for 4 days with vehicle (sesame oil), E 2 (0·05 or 5 µg/rat), P 4 (1 mg/rat) or a combination of both steroids. Rats were included in one of six experimental groups (n=7-8 rats/group): sesame oil (control), low dose of E 2 (0·05 E), high dose of E 2 (5 E), P 4 (P) or combinations 0·05 E+P and 5 E+P.…”

“…OV mice presented only the diestrus pattern while in Sham mice the four stages of the estrous cycle (proestrus, estrus, metestrus, and diestrus) were clearly verified (22). Five days after surgery, all animals were placed on an atherogenic high fat and high cholesterol (HFHC) diet containing 15% fat, 1.25% cholesterol, and 0.5% cholic acid (Cat.…”

“…To obtain pregnant specimens, proestrous females were caged overnight with males with proven fertility. Pregnancy was confirmed by the presence of spermatozoa in the vaginal smear the morning after coitus, defined as day 1 of pregnancy (D1; Freeman 1988, Montes & Luque 1988. In this colony, delivery occurred on D23.…”
