Effects of oestradiol on sex hormone binding globulin

Abstract: Abstract. Sex hormone binding globulin (SHBG) levels were studied for possible effects of oestradiol-17β on SHBG. No change in SHBG plasma was recorded during normal menstrual cycles or during treatment with oestradiol-17β to menopausal women. However, gonadotrophin treatment to amenorrhoeic women to induce ovulation resulted in high oestradiol concentrations and a pronounced increase in SHBG was found during the luteal phase of these cycles. A marked increase of SHBG was also recorded in a woman with pronounc… Show more

“…Our results are consistent with those of previous investigators [1,4,5] but differ from those who reported SHBG levels were increased during the preovulatory and luteal phases of the normal menstrual cycle [6]. The discrepancy regarding SHBG production appears to reflect the individual's metabolic and nutritional status, growth, and aging rather than temporary fluctuations in sex steroids [21], and/or is explained by genetic factors [22,23].…”

“…The plasma concentration of SHBG reflects the balance between estrogen and androgen. No changes in SHBG have been reported to occur during a normal menstrual cycle despite significant increases in preovulatory E 2 levels [1,4,5], however, conflicting findings have been reported [6].…”

“…However, Odlind et at. 26 have observed that there are no cyclical changes in plasma SHBG during the normal menstrual cycle and that extraordinarily high oestradiol levels (150Q-..40(){} pmollL) were required to increase plasma SHBG levels. Clomiphene citrate, an anti-oestrogen, results in a moderate increase in plasma SHBG in men, although this may be mediated by increased plasma oestradiolY Other reported effects of hormones on plasma SHBG levels include elevation of levels following administration of tri-iodothyroninel'' and suppression of elevated SHBG levels by high doses of glucocorticoids in patients with chronic iasthmatic bronchitis?…”

“…and only a markedly elevated level of oestradiol is associated with increased SHBG levels. 26 Therefore, suppressed SHBG levels in obese amenorrhoeic women may be attributable to increased levels of free testosterone, but additional influences cannot be excluded.…”
