Effects of muscle activity and fiber composition on glucose transport and GLUT-4

Abstract: We examined glucose uptake and GLUT-4 in rat muscles [soleus (Sol), plantaris (PL), extensor digitorum longus (EDL), tibialis anterior, and the red and white gastrocnemius (WG)]. In the normally innervated perfused rat hindlimb muscles the proportion of oxidative fibers was highly correlated with the muscle's insulin-stimulated 3-O-methyl-D-glucose (3-MG) uptake (R2 = 0.78) and GLUT-4 content (r = 0.94). Insulin-stimulated 3-MG uptake and GLUT-4 were also highly correlated (R2 = 0.996). In 3-day denervated mus… Show more

“…In addition, the effect of prolonged SB has been demonstrated on muscle GLUT proteins, which are integral for insulin-and exercise-stimulated uptake of plasma glucose. A study of spinal cord injury by Megeney et al found reductions in GLUT content and glucose uptake within 3 days of muscle denervation [28]. Our finding of effect modification by MVPA on the relationship between SB and the components of triacylglycerol, HDL-cholesterol and fasting glucose implies that SB and MVPA may exert their effects through similar pathways for some biomarkers but different pathways for others.…”

“…Similarly, in CD36 null mice, a greater reduction in fatty acid uptake occurred in oxidative muscles than in glycolytic muscles (45). When we examined the effects of reduced muscle activity (denervation) on the changes in glucose transport and transporters (46,47) and lactate transport (48) and monocarboxylate transporters (49), the greatest effects were also observed in the more oxidative types of muscles. Thus, it appears that oxidative types of skeletal muscle are more susceptible to alterations in their substrate transport capacities and transporter expression than glycolytic muscles.…”

“…This effect is temporally coincident with the process of innervation of the muscle fiber (3). Therefore, denervated muscle shows a decrease in GLUT4 and an increase in GLUT1 expression (3)(4)(5)(6)(7). A program of electrical stimulation of the denervated muscle prevents GLUT4 gene repression (3), which suggests that basal contractile activity dependent on innervation regulates the expression of GLUT4 in skeletal muscle.…”

“…A program of electrical stimulation of the denervated muscle prevents GLUT4 gene repression (3), which suggests that basal contractile activity dependent on innervation regulates the expression of GLUT4 in skeletal muscle. Furthermore, GLUT4 is more sensitive than GLUT1 to the lack of muscle contraction (5,8), and the extent of induction of GLUT1 depends mainly on the fiber type (6,7,9).…”
