Effects of Methotrexate on Rabbit Oviducts and on Cell Cultures of Bovine Oviduct Epithelium

Abstract: The influence of a local injection of 0.05 ml/kg weight of 8 mg/ml methotrexate (MTX) was evaluated in 6 rabbit oviducts and compared with saline injections to the contralateral oviducts. Cell cultures of bovine oviducts were incubated in vitro for 7 days and then incubated for 24 h with 100-nmol MTX. Forty-eight and 72 h following the exposure to MTX, the treated and control cultures were harvested. Specimens and cultures were examined by light and scanning electron microscopy. In the in vivo cohort, no diffe… Show more

“…Extrauterine pregnancies would not be such an unusual finding if we were to perform regular necropsies on animals. Different species have successfully been used as animal models in ectopic pregnancy studies, mainly for the evaluation of new treatments and surgical techniques (Nelson et al 1986, Tepper et al 1992, Popp et al 1993) that have later been used in human beings. However, it is necessary to clarify several aspects such as the aetiology of this condition and the implicated factors.…”

“…A model for tubal pregnancy has been established by the arrest of embryos in the Fallopian tubes using appropriately positioned ligatures. Such artificially-retained embryos may progress to the blastocyst stage in the Fallopian tube, but development thereafter is severely compromised and degeneration soon follows (Jacobson et al, 1987;Kooi et al 1992;Lecuru et al, 1992;Popp et al, 1993;Tepper et al, 1992).…”
