Effects of magnesium chloride on in vitro cholinesterase and ATPase poisoning by organophosphate (chlorpyrifos)

Abstract: The present study investigated possible benefits of magnesium ion (as MgCl2) in organophosphorus poisoning targeting its ability to interact with substrates and membrane enzymes. Blood samples collected from volunteered healthy adult by venepuncture into anticoagulant test tubes containing EDTA were separated into plasma and red blood cell and divided into three groups namely: normal, pesticide only (0.25‐2.0 mmol/L chlorpyrifos) and pesticide (0.25‐2.0 mmol/L chlorpyrifos) + 0.1 mol/L MgCl2. Acetylcholinester… Show more

“…Then, the cells were suspended in modified RPMI medium (HyClone, Logan, UT, USA) containing 10% fetal bovine serum (NQBB, Australia) and seeded in 6-well plates (Corning, China). The cells were treated with various concentrations of CPF (20 mg/L, 15 mg/L, 3 mg/L, 1.5 mg/L, 0.75 mg/L, 0.325 mg/L, and 0.1625 mg/L) [27] for 2 h at 25°C in a humidified atmosphere containing 95% air and 5% CO 2 . To determine a suitable working concentration of CPF, cell viability was determined by a Cell Counting Kit-8 (MedChemExpress, Monmouth Junction, NJ, USA) according to the manufacturer's protocols.…”

“…Активность Na + / K + -АТФазы и Са 2+ -АТФазы эритроцитов также была значительно (P < 0,05) снижена хлорпирифосом, в то время как магния хлорид противодействовал эффектам хлорпирифоса со значительным (P < 0,05) повышением активности холинэстеразы, Na + /K + -ATФaзы и Ca 2+ -ATФaзы.
