Effects of Lysozyme on the Activity of Ionic of Fluoroquinolone Species

Abstract: Fluoroquinolones (FQs) constitute an important class of biologically active broad-spectrum antibacterial drugs that are which are in contact with many biological fluids under different acidity conditions. We studied the reactivity of ciprofloxacin (Cpx) and levofloxacin (Lev) and their interaction with lysozyme (Lyz) at different pH values, using UV-visible absorption, fluorescence, infrared spectroscopies supported by DFT calculation and docking. In addition, by antimicrobial assays, the biological consequenc… Show more

“…Furthermore, the drug loading seems to have a negative impact on enzymatic activity and the glucosamine release profiles were higher in 1% loaded systems than in 2% ones. It is reported in the literature that lysozyme interacts with quinolones and this supports that there is a competition between norfloxacin and chitosan, as enzyme substrates, decreasing the enzymatic activity towards chitosan degradation when norfloxacin is at higher concentrations [35]. Moreover, the presence of montmorillonite in the scaffolds could impair lysozyme activity, probably due to a certain degree of interaction between montmorillonite and chitosan, which could prevent chitosan interaction with the enzyme.…”

“…More recently, other fluoroquinolones, in combination with lysozyme, were the subject of DFT calculation and docking, to study their interaction with lysozyme. In 2018, ciprofloxacin and levofloxacin were investigated in antimicrobial assays and an antagonistic effect of lysozyme in an acid medium was evidenced [ 241 ].…”

“…(D) Comparison between the experiment spectra and the ML predictions based on 1000 MD configurations. The black lines represents experiment spectra in amide II region (around 1500–1600 cm –1 ), and the red lines represents the ML predictions spectra based on 1000 MD configurations. Both the ML simulated and experimental spectra were normalized to a uniform scale, where the maximum peak intensity is rescaled to one unit for each protein.…”
