Effects of Long-Chain Fatty Acids on Cytosolic Triacylglycerol Accumulation and Lipid Droplet Formation in Primary Cultured Bovine Mammary Epithelial Cells

Abstract: Mammary epithelial cells have recently been shown to express and secrete leptin into milk and to accumulate triacylglycerol (TAG) in cytosol. We examined the effects on the accumulation of cytosolic TAG of free fatty acid addition to the medium bathing bovine mammary epithelial cells (bMEC). Both saturated (palmitic and stearic) and unsaturated (oleic and linoleic) fatty acids stimulated the accumulation of TAG in a concentration-dependent manner from 50 to 400 microM and the expression of mRNA expression for … Show more

“…In contrast to SCFA, MCFA and LCFA either saturated or unsaturated inhibited leptin expression in bovine adipocytes, in line with those reported in rat and mouse adipocytes [21][22][23][24][25], although they increase leptin expression in bovine mammary epithelial cells [27,28]. The mechanism of the inhibitory effect of LCFA in bovine adipocytes is still obscure, but one plausible explanation is that LCFA acts as a PPAR-γ agonist that has been shown to inhibit leptin expression and secretion in adipocytes [43].…”

“…Similarly, acetate and butyrate stimulate leptin expression in bovine anterior pituitary cells [26]. However, MCFA and LCFA such as caprylate and linoleate, respectively, increase leptin expression in bovine mammary epithelial cells [27,28]. In addition, acetate and butyrate fail to influence leptin expression in bovine mammary epithelial cells [28].…”

“…Elsewhere, bovine PPAR1 and PPAR2 cDNAs have been characterized (Sundvold et al, 1997) with expression of the two isoforms in adipose tissue, whereas only PPARg2 was expressed in the mammary gland. Recently, in primary cultured bovine MEC, the expression of PPARg2 in response to the addition of acetate and octanoate was increased while ACC activity decreased (Yonezawa et al, 2004a), conversely to previous observations in adipose tissue (Lee & Hossner, 2002). Elsewhere, an upregulation of mammary PPAR gene expression was shown in dairy cows between -14 and þ14 days relative to parturition, using a bovine cDNA microarray (Loor et al, 2004c).…”
