“…4 The active metabolite stimulates DA-1 and DA-2 receptors and causes renal and peripheral vasodilatation, and does not seem to have an inotropic effect. 5 Previous studies showed that in patients with heart failure, ibopamine reduced plasma concentrations of noradrenaline, renin activity, and aldosterone. [6][7][8] Clinical studies have reported that ibopamine improves symptoms and has an effect similar to captopril on exercise tolerance in patients with mild-tomoderate heart failure.…”

“…In the presence of the c~-receptor antagonist phentolamine, a relaxant effect of epinine was observed which could be antagonized by the specific D~-receptor antagonist SCH 23390. In contrast to some in vivo studies (Taylor and Cicchetti 1990;Rousseau et al 1992), the precontracted isolated human renal artery rings relaxed only after incubation with the e-adrenoceptor antagonist phentolamine. Also Itoh and colleges (1985) conducted from experiments in dog that a consistent increase in renal blood flow was prevented because the D1 activity of epinine was offset by the ~-adrenergic activity.…”

“…This is in agreement with the in vivo situation. The free plasma concentrations of epinine in vivo have been reported to be similar as the concentration of epinine which produced vasodilatory activity (Rousseau et al 1992). Intravenous administration of epinine at concentrations comparable to those reached after p.o.…”

“…Furthermore, we have shown previously that DI-receptors do not play a major role in the heart, under physiological conditions (Van Woerkens et al, 1991). The lack of a pronounced positive inotropic action of epinine was recently confirmed by infusion of epinine in patients with moderate heart failure due to ischaemic heart disease (Rousseau et al, 1992).…”
