Background: Benign prostate hyperplasia (BPH) is an age-related disease of unknown etiology, characterized by prostate enlargement. The effect of Prosopis africana (PA), Vernonia amydalina (VA) and Ocimum gratissmum (OG), plant extracts on haematological parameters of BPH animal model was investigated. Methods: BPH was induced in 45 male Wistar rats (250-350 g) by exogenous injection of testosterone and estradiol in staggered doses for 3 weeks. To confirm BPH induction, some animals were sacrificed; histological inspection of prostate gland and PSA was carried out. Forty BPH induced rats were divided into 8 groups. Group 1 and 2, 3 and 4, 5 and 6 were treated with 50 mg/kg bw and 100 mg/kg bw doses of PA, VA and OG extracts respectively. Group 7 received finasteride (0.1 mg/kg bw). Group 8 BPH control and five rats without induction constitute group 9, the normal control and both received distilled water. After 45 days, the rats were anaesthetised by a brief exposure to trichloromethane vapour and 5 ml of blood was collected from the rats through cardiac puncture and dispensed into well-labelled EDTA containers to avoid coagulation. All analyses were completed within 24 h of sample collection. Results: Results showed that induction of BPH caused a significant (P< 0.05) enlargement of prostate gland when compared to normal control. All extracts produced significant (P<0.05) reduction in the weight of the enlarged prostate when compared to the BPH control. There were significant (P ˂0.05) decline in RBC, PCV and Hb of BPH control when compared to the normal control and treated groups. In the treated groups the administration of the extracts and standard drug exhibited an increase in RBC, PCV and Hb concentration when compared with the BPH control. Also there was significant (P < 0.05) increase in the WBC, neutrophils, platelets, monocytes, lymphocytes and eosinophils levels in BPH control when compared to normal control and treated groups. In all treated groups there was significant decrease in WBC, neutrophils, platelets, monocytes, lymphocytes and eosinophils concentration levels when compared with the BPH control group. Conclusion: The result of this study indicates that the extracts have the potential to reverse the inflammation caused by BPH and also have the capacity to boost the numbers of red blood cells probably by inhibiting the hemolysis caused by inflammatory factors or by enhancing the production of red blood cell from the bone marrow.

This study was undertaken to evaluate the toxicity effects of root extract of G. kola on serum electrolyte, haematological and histological parameters of rats. Rats of both sexes were randomized into groups and orally administered daily with determined doses of G. kola extract using distilled water as a control for 21 days. On the 22nd day, all the animals were sacrificed and dissected to collect blood and selected organs. The serum and whole blood were assayed for serum electrolytes and haematological parameters respectively while selected organs were examined for their weight and histopathological lesions. The extract of G. kola did not cause significant alteration in majority of the serum electrolytes and hematological indices. However, the extract significantly elevated the mean corpuscular heamoglobin concentration. On the other hand, the extract reduced mean corpuscular volume, haematocrit (150 and 600 mg/kg), mean platelet volume (150 and 600 mg/kg) and procalcitonin (150 mg/kg). In the vital organs, there were no significant lesions observed except at the highest dose. The root extract of G. kola is relatively safe in rats when repetitively administered orally in small doses for a prolonged period of time to ensure that its use is free of toxicity to humans.

Sub-Acute Toxicity Effects of Methanolic Stem Bark Extract of Entada abyssinica on Biochemical, Haematological and Histopathological Parameters in Wistar Albino Rats

Background: Whereas the efficacy of Entada abyssinica (fabaceae) extracts against various ailments has been scientifically validated, its safety has not been established. This study was undertaken to evaluate the toxicity effects of methanolic stem bark extract of E. abyssinica on biochemical, haematological and histological parameters of Wistar albino rats following repeated oral administration.Methods: Wistar albino rats of both sexes were randomized into groups and orally administered daily with determined doses (150, 300 and 600 mg/kg) of E. abyssinica methanolic extract using 1% tween 80 in distilled water as a control for 28 days. On the 29th day, all the animals were sacrificed and dissected to collect blood and selected organs. The serum and whole blood were assayed for biochemical and haematological parameters respectively while selected organs were examined for histopathological lesions. Numerical data was analyzed using graph pad prism and expressed as mean ± standard error of mean. The differences between the treatment and control groups were tested for statistical significance using one-way analysis of variance and/or Student’s t-test.Results: In repeated daily oral doses (150, 300 and 600 mg/kg), the methanolic stem bark extract of E. abyssinica did not cause significant alteration in majority of the biochemical and hematological indices. However, the extract significantly elevated the level of uric acid (all doses), aspartate aminotransferase (300 and 600 mg/kg), low density lipoproteins (150 mg/kg) and mean corpuscular heamoglobin concentration (all doses). On the other hand, the extracts reduced high density lipoproteins (150 and 300 mg/kg), mean corpuscular volume (all doses), haematocrit (150 and 600 mg/kg), mean platelet volume (150 and 600 mg/kg) and procalcitonin (150 mg/kg). In the vital organs, there were no significant lesions observed except at the highest dose (600 mg/kg) where there was mild evidence of lymphocyte infiltration in the liver and focal interstitial nephritis.Conclusion: The methanolic stem bark extract of E. abyssinica is relatively safe in Wistar albino rats when repetitively administered orally in small doses for a prolonged period of time. We recommend more chronic toxicity studies and clinical trials on herbal remedies containing this plant to ensure that its use is free of potential toxicity to humans.
