Effects of Eucommia ulmoides Extract on Longitudinal Bone Growth Rate in Adolescent Female Rats

Abstract: Eucommia ulmoides is one of the popular tonic herbs for the treatment of low back pain and bone fracture and is used in Korean medicine to reinforce muscles and bones. This study was performed to investigate the effects of E. ulmoides extract on longitudinal bone growth rate, growth plate height, and the expressions of bone morphogenetic protein 2 (BMP-2) and insulin-like growth factor 1 (IGF-1) in adolescent female rats. In two groups, we administered a twice-daily dosage of E. ulmoides extract (at 30 and 100… Show more

“…P. umbrosa at doses of 100 and 300 mg/kg was shown to increase the bone growth rate to 380.9 and 381.5 μm/day in the proximal tibial growth plate, respectively. The bone growth rate in the control group was 358.2 μm/day in accordance with the results reported previously [41]. Tetracycline gets deposited in newly formed bones, causing a fluorescent line corresponding to the injection, and the distance between the chondro-osseous junction and the fluorescent line indicates the growth rate, which is the length of bone growth during a certain period of time [37].…”

“…Finally, the sections were photographed with a microscope. The percentage of labelled chondrocytes was calculated by counting stained and total chondrocytes in two parallel columns using Image J software [ 3 ].…”

“…The growth plate consists of three distinct zones: the resting, proliferative, and hypertrophic zones. Chondrocytes in the growth plate are differentiated from the top of the resting zone to the bottom of the hypertrophic zone, and the middle proliferative zone is the major location for chondrocyte proliferation for bone elongation [ 2 , 3 ]. Growth and differentiation of the growth plate are regulated by growth hormone (GH) and cell-signalling polypeptides such as insulin-like growth factor (IGF)-1 and bone morphogenetic protein (BMP)-2 [ 4 ].…”

“…In ovariectomized rats fed with Eucommiae Cortex extract, the femur biomechanical quality and trabecular microarchitecture were improved without hyperplastic effect on uterus [234]. When adolescent female rats were treated with the bark extract, promotion of longitudinal bone growth was observed, with increased BMP-2 and IGF-1 expressions in the proliferative and hypertrophic zones [235]. In a disuse-induced osteoporosis model of hind-limb suspended rats, the Eucommia bark extract could prevent bone loss as indicated by decreased levels of bone turnover markers; it enhanced the biomechanical bone strength and prevented the deterioration of trabecular bone microarchitecture [236].…”
