Effects of hypertonic saline solution in inflammatory response in sepsis

Abstract

Sepsis is a systemic inflammatory response to infection and a major cause of mortality. The hypertonic saline solution (HSS) has been showed to improve hemodynamic and inflammatory response. We studied the hypertonic solution effects in sepsis through cecal ligation and puncture (CLP) in mice Balb‐C, divided in 3 groups: CLP without treatment (C); CLP treated with saline solution 0,9% ‐ 34mg/Kg (SS); CLP treated with HSS 7,5% ‐ 4ml/Kg (SH); and a 4° group to indicate the basal values without both CLP and treatment; both treatments (HSS and saline) were applied 30 min after CLP. The lung was collected after 6, 12 e 24 h to analyze cytokines levels by ELISA and nitric oxide (NO) by Griess method. The group SH showed higher survival rates (60%) when compared to SS (46,6%) and C (33,3%). The SS group (27,3 ± 3,05 pg/ml) presented increased TNF‐α level in 12 h when compared to group C (14,07 ± 1,68 pg/ml) and SH (19,66±3,19 pg/ml). After 24 h the SH (6,2 ± 3,5 pg/ml) decreased TNF‐α when compared to C (73,40± 49,52 pg/ml); IL‐10 level in 24 h was increased in C (CLP ‐ 3,12 ± 0,46 pg/ml) and SS (2,9 ± 0,9 pg/ml) in relation to SH (0,59 ± 0,34 pg/ml). NO concentration was lower in SH (2,26 ± 1,34 pg/ml) compared with C (10,96 ± 2,2 pg/ml) after 24 h. The HSS protects the animal against sepsis, but saline solution doesn't. Our results suggest that the volume replacement of both treatments modulate pro and anti‐inflammatory cytokines and NO. CAPES.FAPESP.
