Effects of histamine type-2 receptor antagonists on indomethacin and IL-2 immunotherapy of metastasis

Abstract: Histamine type-2 receptor antagonists (H-2RA) have been used chronically to prevent dyspepsia in cancer patients subjected to immunotherapy with chronic indomethacin (Indo) and intermittent IL-2 in our cancer centre. We tested the effects of these agents during immunotherapy of C3H/HeJ mice transplanted s.c. with 5 x 10(5) C3L5 mammary adenocarcinoma cells. Tumor-transplanted mice were divided into groups receiving: (1) Indo (14 micrograms/ml); (2) H-2RA, i.e. (a) ranitidine at 28.6 micrograms/ml (Ran-lo) or 1… Show more

“…C3L5, a highly metastatic murine mammary tumor cell line was obtained from Dr. P. K. Lala (University of Western Ontario, Canada). This cell line was generated by five cycles of repeated in vivo selections for spontaneous lung micrometastases following subcutaneous transplantation of the primary line derived from a spontaneous mammary adenocarcinoma in female retired breeder C3H/HeJ mouse (Lala and Parhar, 1993; Saarloos et al, 1993; Jadeski and Lala, 1999; Timoshenko et al, 2004). Each of the breast cancer cell lines was maintained in RPMI 1640 containing penicillin (100 U/ml), streptomycin (100 U/ml), and 10% FBS.…”
