Effects of Herbicides on RNA and Protein Syntheses

Abstract: Effects of 22 herbicides on the synthesis of RNA and protein were investigated in excised tissues. The assays measured ATP and orotate incorporation into RNA, leucine incorporation into protein, and the gibberellin-controlled induction of a-amylase Statistical analysis of the average responses measured in the four assays suggested that 14 of the herbicides inhibited RNA and protein biosynthesesin vivo. The most inhibitory chemicals were 4-hydroxy-3,5-diiodobenzonitrile (ioxynil), 2-sec-butyl-4,6-dinitrophenol (… Show more

“…Results reported herein for both intact and excised root tips indicate that the initial effects of trifluralin are not on protein biosynthesis . This is in agreement with reports of studies with excised soybean hypocotyls [5] and corn mesocotyl and soybean hypocotyl sections [14] . However, if the synthesis of a specific protein was affected, the techniques used in this study would not detect changes at that level .…”

“…Moreland et al . [14] found no measurable effect on ATP generation, orotate incorporation into RNA or leucine incorporation into protein in maize mesocotyl or soybean (Glycine max L.) hypocotyl sections . Protein synthesis (14C-leucine incorporation) in tobacco callus was inhibited, but 5 to 7 days after treatment [27] .…”

“…A single one-halfcentimeter shoot segment immediately above the seed and a single 1.0-cm root tip segment were excised from the seedlings and kept at 4~ until a sufficient amount had been obtained. The incubation medium used for this study contained 0.01 M KHzPO 4 buffer (pH 6.0) containing 1% sucrose (w/v) and 40 IxM chloramphenicol (Moreland et al 1969).…”

“…Streptomycin, at the concentration used in these experiments, has been reported to control bacterial growth without seriously affecting precursor incorporation into RNA and protein of higher plant tissues (8). Incorporation of a labeled nucleotide such as 14C-ATP into RNA and of a labeled amino acid such as 14C-leucine into a polypeptide chain has been widely used to measure and study de novo RNA and protein synthesis in a variety of plant tissues (8,14).…”
