In the past 30 years, many amphibian species have suffered population declines throughout the world. Mass mortality have been frequently reported, and in several instances, infectious diseases appear to be the cause of death. The role that contaminants could play in these die-offs through immunotoxic effects has been poorly investigated. In this study, juvenile leopard frogs (Rana pipiens) were exposed for 21 d to a mixture of six pesticides (atrazine, metribuzin, aldicarb, endosulfane, lindane, and dieldrin) and subsequently challenged with a parasitic nematode, Rhabdias ranae. Exposure to the mixture at environmentally realistic concentrations significantly reduced lymphocyte proliferation. Three weeks after the end of the exposure, lymphocyte proliferation had recovered and was stimulated in frogs challenged with parasites with the exception of those previously exposed to the highest concentration. No pesticide effects on phagocytosis and splenocyte numbers were detectable at the end of the exposure period, but these two parameters were diminished 21 d after the infection challenge in frogs previously exposed to the highest levels of pesticides. In these animals, the prevalence of lung infection by R. ranae also tended to be higher. These results suggest that agricultural pesticides can alter the immune response of frogs and affect their ability to deal with parasitic infection.

Other studies conducted with natural exposure or oral administration of trace elements [39,40] showed that increasing concentrations of some metals (Hg, Cu, Pb, Cd, Zn) increased the number of the monoclonal parasite nematodes Ascaris suum (Goeze, 1782) in guinea pigs Cavia porcellus (Pallas, 1766), as the result of a considerable suppression of lymphocytes T and B and of the phagocytic ability of macrophages during the migration phase of Ascaris suum, when compared to a group of infected animals that remained untreated. Parasitological, hematological, and ecotoxicological analyses were carried out on a population of lesser spotted dogfish Scyliorhinus canicula from the central Mediterranean Sea. Parasitological analyses highlighted a poor helminthic community, highly dominated by a single taxon represented by the cestode Nybelinia sp. No differences in the parasitic load between females and males were observed. Hematological analyses showed that the number of leukocytes was significantly lower in the sharks that resulted in parasitism, and this could be due to the ability of some trace elements, such as arsenic, weakening the immune system and exposing animals to a higher risk of parasite infection, although further hematological and parasitological analyses are required on a larger number of samples. Trace elements analyses in the vertebrae, skin, and liver highlighted that the most abundant and potentially toxic elements were lead (Pb), arsenic (As), and cadmium (Cd). Other trace elements were also abundant, such as manganese (Mn), zinc (Zn), nickel (Ni), copper (Cu), and iron (Fe). Pb, As, and Mn showed the highest concentrations in vertebrae, while Cd, Cu, and Zn were the highest in the liver, probably due to their concentration in the prey items of the sharks; Fe and Ni showed the highest concentrations in the skin, due to their presence in the water column, especially along the coast where animals were collected. The concentration of some trace elements analyzed in the vertebrae decreased with the growth of the sharks. These results confirm that elasmobranchs, being predators at the apex of the marine food chain, act as final receptors for a series of polluting elements regularly discharged into the sea.

The subchronic effect of mercury on selected immunological parameters was studied in guinea pigs with experimentalAscaris suuminfection. HgCl2given for 28 days reduced significantly T- and B-cell populations in the lymphoid organs and the phagocytic ability of peritoneal macrophages. The subsequent infection of HgCl2-intoxicated animals elevated the studied immunological parameters, but in comparison with infected non-intoxicated guinea pigs they remained significantly suppressed. The mercury compound in infection stressed animals caused a slight alteration of the complement CH50and AH50activity. The specific circulating antibody level in infected and HgCl2treated animals rose a little by day 12 p.i. and then again decreased significantly, compared with untreated guinea pigs. The mean intensity of infection with migratingAscarislarvae in HgC12-treated animals increased by 15%, compared with controls.
