Effects of growth hormone therapy in prepubertal children with short stature secondary to intrauterine growth retardation

Abstract: A total of 130 short children were included in a French multicentre study and randomized between a control group (group A) and two groups treated with daily subcutaneous injections of GH at doses of 0.7 IU/kg/week (group B) and 1.4 IU/kg/week (group C) for 2 years. Height velocity was significantly increased (p<0.0005) in groups B and C, with a greater increase in group C than in group B (p< 0.001). The benefit after 2 years compared with controls was 4.3 cm in group B and 5.9 cm in group C. The rate of bone m… Show more

“…GH was started at a mean age of 7.5 years. Five years of GH therapy at a dose of 1 vs. 2 mg/m 2 per day resulted in a significant improvement in mean (SD) height SDS of 2.12 (0.52) vs. 2.57 (0.72), respectively, and a significant increase in mean (SD) height SDS for bone age of 1.06 (0.78) vs. 1.40 (1.37), respectively [17]. After 5 years of GH therapy the mean gain in height SDS was statistically just significantly greater in children treated with the higher GH dose of 2 mg/m² per day.…”

“…In a randomized, double-blind, dose-response GH trial 79 Dutch short SGA children were continuously treated until adult height with either 1 or 2 mg GH/m 2 per day [16, 17]. Children with chromosomal abnormalities, syndromes (except Silver-Russell syndrome) and severe dysmorphic symptoms were not included.…”

“…Initial data were disappointing, probably due to the low dose and frequency of GH administration. Recent short term studies have shown that daily administration of recombinant human GH therapy in varying dosages accelerates growth significantly in short children born SGA (7,(15)(16)(17)(18)(19)(20)(21)(22).…”

“…(27)(28)(29)(30)(31)(32)(33). O efeito dose-dependente nos primeiros anos de tratamento se tornou evidente (29,30,34,35). Um ganho médio na altura de 1,72 DP foi demonstrado após três anos de tratamento nas crianças recebendo 0,1 UI/kg/dia de GH, comparado com um ganho médio de 2,54 DP nas crianças recebendo 0,2 UI/kg/dia.…”
