Effects of growth hormone and thyroxine on kidney insulin-like growth factor-I and renal growth in hypophysectomized rats

Abstract: The effects of treatment for 11 days with human growth hormone (hGH; 140 micrograms/day), thyroxine (T4; micrograms/day) and hGH+T4 on renal growth and content of insulin-like growth factor-I (IGF-I) in hypophysectomized rats have been compared with saline-treated hypophysectomized animals and intact control animals. “…It thus seems clear that although renal IGF-I mRNA accumulation takes place both in hypophysectomized untreated and p-UC-UBI-hGH and pEGFP-N1-treated mice, the regulation may be essentially different, theoretically involving enhanced non-transcriptional and/or post-transcriptional mechanisms. Previously it was shown that treatment with physiological doses of thyroxin (T4) stimulated and increased renal weight, and it was suggested that renal growth is not primarily mediated through IGF-I, but by an alternative mechanism [13]. It has previously been demonstrated that treatment of hypophysectomized animals with similar doses of T4 normalizes the renal content of T3 and the activity of the iodothyronine deiodase, whereas hGH treatment has little effect on these parameters [8].…”
