Effects of gossypol on the motility of mammalian sperm

Abstract: The effects of the male contraceptive gossypol on the motility of mammalian spermatozoa are reviewed. The role of sperm motility in the processes of fertilization and the effect of the drug on these processes determine its effectiveness as a contraceptive. The promising male contraceptive potential of gossypol is discussed in the context of the serious adverse effects of the agent.

“…The adverse effects of SAL on the testis are comparable with that of polyphenols like gossypol. Similar dysfunctions in the testis including decreased weight, regressed seminiferous tubules, reduced number of spermatozoa with reduced motility were reported in many mammalian species following gossypol administration [5254]. The vacuolated epithelium of epididymis and aggregation of necrotic cells in its lumen indicated abnormal spermatogenesis as well as lack of testosterone supply [55].…”

“…It has been used traditionally to treat bronchitis and to induce abortion and menses, but was only recognized as a male antifertility agent in the 1970s. [40] Whether infertility induced by gossypol is reversible is in dispute. GOS is toxic to non-ruminant animals and has limited the use of cottonseed meal as a dietary source of proteins for farm animals.…”

“…[28] Studies on the mechanism of action of gossypol suggest structural alterations of cell membranes, inhibition of enzymes and energy metabolism may be affected. [40] The effect of GOS on spermatogenesis results in the arrest of spermatozoa development and finally sterilization of male. [50] Deleterious effects of GOS toward spermatozoa include immobilization of spermatozoa motility and fertilizing ability.…”
