Effects of two maturation media with and without the addition of oestrus buffalo serum (OBS) and hormones (FSH, LH, E2) on the maturation rate of buffalo follicular oocytes were evaluated. The results revealed a signifi cant (P < 0.05) increase in the maturation rate when the OBS and hormones were added to TCM-199 than in Ham's F-12 medium. The percentage of maturation rates in TCM-199 + hormones + OBS, 55.17, 62.28 and 26.62 Reproduction, Bubalus bubalis, TCM-199, Ham's F-12, FSH, LH, E2Riverine buffaloes (Bubalus bubalis) are hardy dairy animals, resistant to climate, stress and diseases. However, problems like delayed onset of reproductive maturity, seasonality of breeding, long calving interval, latent oestrus, low number of primordial follicles and poor superovulatory response have been attributed to poor reproductive performance of this species (Nandi et al. 2002).In vitro maturation (IVM) and in vitro fertilization (IVF) procedures performed on oocytes obtained from slaughter-house derived ovaries have recently provided a practical means for producing large number of bovine zygotes at low cost for research and commercial settings (Hansen 2006). Application of this technology in assisted reproduction of buffalo will not only improve productive and reproductive potential of the buffalo population but will also help rescue the precious germ plasma going to waste by indiscriminate slaughter of this animal.The acquisition of more insights into the buffalo culture requirements is critical to optimize the effi ciency of advanced reproductive strategies in this species. Lack of proper conditions to support in vitro culture of buffalo zygotes to a transferable stage embryo is the major impediment in the successful buffalo IVF system. The direct extrapolation of the methods from cattle to the buffalo resulted in poor fertility. Chauhan et al. (1997) reported that oocyte maturation and cleavage rates in buffalo are lower than those for cattle. There have been reports to enhance early development of in vitro produced embryos in domestic animals by addition of hormones, sera and somatic cells (Pawshe et al. 1996). To optimize in vitro embryo production of buffalo by improving the culture conditions, the present project was designed: 1) to assess the effect of addition of sera and hormones on in vitro maturation of buffalo oocytes and 2) to evaluate media in supporting fertilization of in vitro matured oocytes.

Summary:The present study was aimed at the investigation of the effects of Recombinant Follicle Stimulating Hormone (rFSH) on fertility in Awassi ewes synchronized with PGF 2 α in the breeding season. The fifty thoroughbred Awassi ewes used in the study were injected twice with 0.150 mg of Tiaprost (PGF 2 α, Iliren®) at a 10-day interval. Following the second PGF 2 α injection, the ewes were randomly distributed into two equal groups. The ewes included in the first group were not subjected to any additional injection and were referred to as the PGF 2 α group (Group 1, n=25). The animals included in the second group (Group 2, n=25) were administered 10 IU Follitropin (rFSH, Puregon ® ), diluted 1:10 with physiological saline, 24 hours before the second PGF 2 α injection. Ewes were tested for estrus by a fertile teasing ram three times a day (06.00, 14.00 and 20.00 h), for one hour each time. Ewes exhibiting estrus were mated naturally. Pregnancy was determined by transrectal ultrasonography on 40. d after mating. Estrus was observed at a rate of 88.0 % in Group 1 and 92.0 % in Group 2. The rate of pregnancy was 100 % in the first group and 95.6% in the second group. The rate of lambing was 100 % and 95.5 % in Groups 1 and 2, respectively. Furthermore, twin pregnancy rates were 4.5 % in the first group and 57.1% in the second group. While differences between the groups were determined to be significant (p<0.001), no significant difference was detected for pregnancy, estrus or lambing rates. In conclusion, it was determined that the administration of rFSH to Awassi ewes in the breeding season, 24 hours before the second PGF 2 α injection, led to an increase in twin pregnancy rates. Thereby, this protocol can be used as an alternative synchronization method to increase fertility. It has been concluded that further studies are needed to determine the effect of different doses of rFSH on fertility in ewes.Key words: Awassi ewes, fertility, PGF 2α, rFSHAşım sezonunda PGF 2 α ile senkronize edilen İvesi koyunlarında bazı fertilite parametreleri üzerine rekombinant follikül stimüle edici hormonun (rFSH) etkisi Özet: Sunulan çalışmanın amacı, aşım sezonunda PGF 2 α ile senkronize edilmiş İvesi Koyunlarında Recombinant Follikül Stimüle Hormonun (rFSH) fertilite üzerine etkisi araştırmaktır. İkinci PGF 2 α enjeksiyonları yapıldıktan sonra koyunlar rastgele iki eşit gruba ayrıldı. Birinci gruptaki koyunlara ilave bir enjeksiyon yapılmadan (Grup 1, n=25) PGF 2 α grubu, ikinci gruptaki (Grup 2, n=25) koyunlara ise ikinci PGF 2 α enjeksiyondan 24 saat önce 1/10 oranında serum fizyolojik ile sulandırılıp 10 IU Follitropin (rFSH, Puregon ® ) enjeksiyonu yapıldı. Summary Preparation of oocytes is one of the critical factors that determine the developmental competence of embryos produced by in vitro fertilization (IVF). The present study was conducted to evaluate the effect of different maturation media on the nuclear and cytoplasmic maturation of buffalo oocytes. Also, to investigate if the addition of ginseng and fenugreek to the maturation media during in vitro maturation (IVM) would influence on IVM, and developmental competence of buffalo oocytes as well as intracellular GSH level. Three experiments were conducted to achieve the goals of present study. In the first experiment, the buffalo oocytes were cultured in different media without any supplementations to detect the most suitable effective medium for IVM. The second experiment compared between different culture media for IVM of oocytes through measurement of intracellular glutathione level. The third experiment examined the effect of ginseng and fenugreek additives during the IVM of Egyptian buffalo oocytes. The results of this study proved that the TCM-199 medium significantly increased the percentage of cumulus expansion, nuclear maturation and intracellular GSH concentration after IVM of buffalo oocytes, while CR1aa was the least effective medium. Treatment with ginseng and fenugreek extracts during IVM culture efficiently improves the development competence of buffalo oocytes, and this improvement is correlated with increase of intracellular GSH concentration. In conclusion, TCM-199 was the most effective medium and is desirable for in vitro maturation of Egyptian buffalo oocytes. Ginseng and fenugreek additives enhanced the quality of less effective medium (CR1aa), as well as optimized the effect of TCM-199 medium.
