Effects of glutaraldehyde fixative osmolarities on smooth muscle cell volume, and osmotic reactivity of the cells after fixation

Abstract: The contribution of glutaraldehyde (GA) to the effective osmolarity of GA fixatives, the osmotic reactivity of the cells after fixation in GAY and also the duration of fixation in GA on cell volume, were investigated using cultured smooth muscle cells (SMC) and spiral aortic strips. Four fixation procedures were studied. We found that GA contributes to the total effective osmolarity of the fixatives, and that the type of buffers used for the fixatives can also affect the cell volume differently during GA fixat… Show more

“…Hillman and Deutsch (1978) reported a 20-30% decrease in tissue cross-sectional areas of rat brain with glutaraldehyde fixation and ethanol dehydration. Similarly, cell volume changes of 10-30% were reported in cultured rat smooth muscle tissue (Lee et al, 1982). These area and volume differences related to chemical fixation need to be understood and minimized so that comparisons between fixed tissue and living tissue may be made.…”

“…Moreover, Lee and co-workers reported that hyperosmotic medium led to contractions of VSMR after their fixation with glutaraldehyde [44]. It should be underlined, however, that despite the different mechanisms of force generation in shrunken and swollen SMC, their contractions were equally sensitive to actin cytoskeleton disruption.…”
