Effects of gamma irradiation and periodate oxidation on the structure of dextrin assessed by mass spectrometry

Abstract: A B S T R A C TSterilization of biomaterials by gamma irradiation should not change its structure, since this may affect also its bioactivity. In this work, dextrin and periodate-oxidized dextrin (ODEX), both irradiated and non-irradiated, were hydrolysed by α-amylase and the obtained oligosaccharides were fractionated by ligand-exchange/size exclusion chromatography and analyzed by matrix-assisted laser desorption/ionization (MALDI-MS) and electrospray ionization (ESI-MS n ) mass spectrometry. This allowed th… Show more

“…This is the case of dextran 67,68 and hydroxyethyl starch, 69,70 both used as plasma volume expanders. 25 The obtained results suggest that such modification did not promote any skin sensitization. 36,37 It is generally safe and well tolerated by PD patients, but there are few reports of acute self-limiting allergic hypersensitivity reaction to icodextrin, such as skin rashes.…”

“…Dextrin oxidation was performed as previously described by Pereira et al 25 Briefly, aqueous solutions of dextrin (2% w/v) were oxidized with sodium m-periodate, to yield the theoretical degree of oxidation of 40%, at room temperature, with stirring, in the dark. “…The formation of DCS is confirmed by HRMS spectral analysis of dextrin (DEX) and DCS where the corresponding figures are shown in S1 and S2 respectively. The Figure S1 (a, b) shows that the peaks at m/z =169.04 and 203.00 correspond to the [C 6 H 10 O 4 +Na] + and [C 6 H 12 O 6 +Na] + moieties respectively, indicating the presence of anhydrous glucose unit (AGU) in dextrin polysaccharide . However the Figure S2 (a, b) shows that the new peaks appear at m/z = 133.02 and 107.04 that correspond to [C 4 H 6 NO 2 S+H] + and [C 3 H 6 O 2 S+H] + units.…”
