Effects of ethanol on psychomotor performance under steady-state conditions

Abstract: Ethanol was administered to eight male volunteers using an oral loading dose followed by repeated small oral doses to achieve approximate steady-state drug concentrations in a double-blind placebo controlled cross over design. Ethanol or placebo were administered over a 5-h period in two sessions at least 7 days apart. The effects of ethanol were assessed using a short battery of psychomotor tests and visual analogue scales which was administered repeatedly during the steady-state period, and a long battery ad… Show more

“…This review identified seven studies that used a steady-state paradigm and five found evidence of acute behavioral tolerance. Two studies maintained a constant BAC by giving an initial oral dose to reach the desired BAC and then repeatedly giving oral maintenance doses throughout, but neither reported acute tolerance occurring on any measure (Fagan, Tiplady, & Scott, 1994; Kaplan, Sellers, Hamilton, Naranjo, & Dorian, 1985). The remaining five studies using the steady-state paradigm maintained a constant BAC through means of an intravenous clamp.…”

“…Others have attempted to demonstrate the development of acute tolerance to alcohol while BrAC was held constant (Fagan et al, 1994;Hiltunen et al, 2000;Kaplan et al, 1985). Approximate steady-state BrACs (albeit with high variance) were achieved in social drinkers with either intravenous or repeated oral administration of alcohol.…”

“…Maze tests seem to be much less commonly used than they were before the advent of personal computers, although they are sensitive measures of drug effects (Frewer and Lader, 1993;Fagan et al, 1994). This may be related to the inconvenience of using paper-based tasks compared to tasks that can readily be automated.…”
