The overproduction of specialized metabolites requires plants to manage the inherent burdens, including the risk of selfintoxication. We present a control mechanism that stops the expression of phytoalexin biosynthetic enzymes by blocking the antecedent signal transduction cascade. Cultured cells of Eschscholzia californica (Papaveraceae) and Catharanthus roseus (Apocynaceae) overproduce benzophenanthridine alkaloids and monoterpenoid indole alkaloids, respectively, in response to microbial elicitors. In both plants, an elicitor-responsive phospholipase A2 (PLA2) at the plasma membrane generates signal molecules that initiate the induction of biosynthetic enzymes. The final alkaloids produced in the respective plant inhibit the respective PLA, a negative feedback that prevents continuous overexpression. The selective inhibition by alkaloids from the class produced in the "self" plant could be transferred to leaves of Nicotiana benthamiana via recombinant expression of PLA2. The 3D homology model of each PLA2 displays a binding pocket that specifically accommodates alkaloids of the class produced by the same plant, but not of the other class; for example, C. roseus PLA2 only accommodates C. roseus alkaloids. The interaction energies of docked alkaloids correlate with their selective inhibition of PLA2 activity. The existence in two evolutionary distant plants of phospholipases A2 that discriminate "self-made" from "foreign" alkaloids reveals molecular fingerprints left in signal enzymes during the evolution of species-specific, cytotoxic phytoalexins.

A comprehensive metabolomic profiling of Catharanthus roseus L. G. Don infected by 10 types of phytoplasmas was carried out using one-dimensional and two-dimensional NMR spectroscopy followed by principal component analysis (PCA), an unsupervised clustering method requiring no knowledge of the data set and used to reduce the dimensionality of multivariate data while preserving most of the variance within it. With a combination of these techniques, we were able to identify those metabolites that were present in different levels in phytoplasma-infected C. roseus leaves than in healthy ones. The infection by phytoplasma in C. roseus leaves causes an increase of metabolites related to the biosynthetic pathways of phenylpropanoids or terpenoid indole alkaloids: chlorogenic acid, loganic acid, secologanin, and vindoline. Furthermore, higher abundance of Glc, Glu, polyphenols, succinic acid, and Suc were detected in the phytoplasma-infected leaves. The PCA of the 1 H-NMR signals of healthy and phytoplasma-infected C. roseus leaves shows that these metabolites are major discriminating factors to characterize the phytoplasma-infected C. roseus leaves from healthy ones. Based on the NMR and PCA analysis, it might be suggested that the biosynthetic pathway of terpenoid indole alkaloids, together with that of phenylpropanoids, is stimulated by the infection of phytoplasma.

SUMMARY 7Often when a project is initiated plant cell cultures produce low levels of a desired compound. For the project to move to a commercial level, time-dependent volumetric yields (mg.product/L-day) must typically be increased several orders of magnitude. Typically, no single growth enhancing strategy will give such a large increase, but in many cases the simultaneous application of multiple-strategies results in a synergistic interaction and the large increases necessary to bring yields in a plant cell culture process to a useful level. Yield improvement strategies include selection of stable, high-yielding variants; manipUlation of medium components and/or gas phase composition; metabolic manipulation through gene manipulation or use of inhibitors; precursor feeding; use of elicitors; in situ product removal; and use of immobilization or more differentiated or organized plant material. INTRODUCTIONThe commercial application of plant ceIl cultures is likely to be in three areas: (1) facile production of elite plants; (2) as a tool to generate sufficient quantities of metabolites for screening or characterization in a drug discovery program; and (3) for the commercial scale production of a metabolite. In this symposium and book we are focused on the third objective, particularly as it applies to food ingredients. Reviews on food ingredient production and a possible role for plant cell cultures has been published elsewhere
