Effects of Direct Hemoperfusion with a β2-Microglobulin Adsorption Column on Hypercytokinemia in Rats

Abstract: Background/Aim: Blood purification therapies have been clinically applied to treat the cytokine-induced pathological responses. The effects of direct hemoperfusion(DHP) with a β2-microglobulin (BMG) adsorption column on hypercytokinemia were investigated. Methods: A hypercytokinemia model was prepared by injecting lipopolysaccharide (LPS) into femoral veins of male Wistar rats weighing 250–400 g. The hypercytokinemic rats were connected to the DHP system using a small BMG adsorption column, and time… Show more

“…LixelleS‐35 is a β 2 MG adsorption column that consists of porous cellulose beads (18), has a ligand of the hexadecyl group on the surface, and is able to adsorb β 2 MG by hydrophobic interaction. It also adsorbs various cytokines (19). In healthy subjects, the concentration of kappa FLC ranges from 3.3 to 19.4 mg/L and that of lambda FLC ranges from 5.7 to 26.3 mg/L.…”

“…TNF‐α concentrations were 3270 ± 657 and 6198 ± 640 pg/mL 60 min after LPS injection, 5831 ± 494 and 9037 ± 317 pg/mL 90 min after LPS injection, 5902 ± 463 and 8127 ± 374 pg/mL 120 min after LPS injection, and 3613 ± 254 and 5408 ± 317 pg/mL 150 min after LPS injection in the adsorbent and control groups, respectively. Thus, the increase in TNF‐α concentration was significantly inhibited in the adsorbed group at the time points indicated ( P < 0.05) (16).…”
