Effects of Different Growth Media on In Vitro Seedling Development of an Endangered Orchid Species Sedirea japonica

Abstract: Sedirea japonica is becoming endangered, and even extinct, due to habitat destruction and illegal collection, and the development of an optimized artificial propagation system is necessary for its conservation and reintroduction. Thus, the effects of plant growth medium strength (Murashige and Skoog (MS) and Hyponex media) and the addition of activated charcoal (AC) and organic supplements on seedling growth of S. japonica were investigated through in vitro seed culture. The results showed that seedling growth… Show more

“…On the other hand, the use of Half-MS strength medium also resulted in the best growth and development of PLBs in Dendrobium var. Sonia [6], Cattleya bicolor orchid [7], Sedirea japonica orchid [13], and Dendrobium seedlings [10]. The use of Half-MS strength medium also proves that for the growth and development of PLBs, several types of orchids do not require high concentrations.…”

“…Enrichment with 0.6 gL -1 coconut water, 30 gL -1 banana homogenate, and 10 gL -1 apple-homogenate into MS medium very effectively stimulated the regeneration of orchid PLBs Sedirea japonica and increasing plantlet fresh weight. The results of this study have been standardized as a medium for mass production and conservation of this rare epiphytic orchid [13,14]. The use medium NP + 100 gL -1 banana in in vitro culture media increases the growth rate of D. lineale orchids [15].…”

“…The promoting effect of the Hyponex medium on the GGB proliferation and differentiation might be correlated to the relatively high content of trace elements and microelements in the medium. Despite the wide application of Hyponex in the tissue culture of orchids (An et al, 2021;Chen et al, 2015;Ket et al, 2004), there have been no reports on the use of the Hyponex medium in the tissue culture of ferns.…”

“…As demonstrated in previous studies, incorporating organic supplements into culture media can enhance the growth and development of various plant tissues in vitro. These supplements include apple juice, banana homogenates, coconut water, corn extracts, and potato homogenates [34,35]. These organic additives are integrated into the culture medium not only as a natural source of carbon, but also for their rich content in vitamins, fiber, hormones, and proteins [36].…”
