Effects of dietary salt changes on renal renin-angiotensin system in rats

Abstract: Renin (RA) and angiotensin-converting enzyme (ACE) activities and angiotensinogen, ANG I, and ANG II levels were measured in the kidney (cortex and medulla) and plasma of Wistar-Kyoto rats on a low-sodium (LS; 0.025% NaCl; n = 8), normal-sodium (NS; 1% NaCl; n = 7), or high-sodium (HS; 8% NaCl; n = 7) diet for 21 days. RA, ANG I, and ANG II levels increased in a manner inversely related to sodium content of the diet in both plasma and renal tissues. The LS diet resulted in a 16-, 2.8-, and 1.8-fold increase in… Show more

“…Renin is the rate-limiting step in the production of ANG II through its conversion of angiotensinogen into angiotensin I (22) and therefore gives an indication of overall endogenous RAS activity. The current study has demonstrated that plasma renin concentration was augmented in low-sodium-fed rats and markedly reduced in high-sodium-fed rats, and these data concur with findings from other groups (9,14). Furthermore, previous studies have shown both circulating (8,15) and intrarenal (9,14) ANG II levels were also increased in lowsodium-fed rats and reduced in high-sodium-fed rats.…”

“…This was examined by 1) determining the impact of intrarenal infusion of ANG (1-7) on renal hemodynamic and excretory function; and 2) investigating how placing animals on a low-and high-sodium diet, to activate or suppress the endogenous RAS (9,14), would alter the hemodynamic and excretory actions of ANG (1)(2)(3)(4)(5)(6)(7) Surgical protocol. Rats were fasted overnight but did have access to water.…”

“…This involves endogenous Ang II as an effector of proximal reabsorption and its modulation by dietary salt. The reciprocal effects of dietary salt on plasma and kidney Ang II in the present studies were entirely expected, since we have previously documented that plasma and whole-kidney concentrations of Ang II increase during dietary salt restriction in rats (20), and others have confirmed this (21). Therefore, we surmised that Ang II would exert a lesser tonic influence over proximal reabsorption in rats fed a high-salt diet and that this would appear as a lesser decrease in proximal reabsorption during treatment with losartan.…”
