Semen quality has certainly declined over the past few decades, possibly owing to modern lifestyle factors. In this sense, the role of overweight and obesity in the development of subfertility in males has generated a considerable amount of interest in recent years. However, there is no consensus on whether overweight or obesity impaired sperm quality. Thus, based on the ongoing debate about risk factors for subfertility associated with overweight and obesity in men, this study was designed to investigate the effect of overweight on sperm quality parameters and fertility success in randomized controlled trial in a rabbit model. Fourteen male rabbits were randomly assigned to a control group in which nutritional requirements were satisfied or a group fed to satiety from 12 to 32 weeks of age. At 24 weeks of age, semen samples were analysed weekly by conventional semen analysis for 8 weeks. In addition, during the trial female rabbits were artificially inseminated by each male to assess the fertility success and the number of offspring. Young males fed to satiety were associated with a significant increase in body weight (13.6% overweight) and perirenal fat thickness (5%). Male overweight presented a significant decrease in sperm concentration. There were no differences in the remaining sperm parameters. However, male overweight showed a clear and significant decrease in fertility success (control group, 64±8.9% versus fed to satiety group, 35±9.2%), but not in the number of offspring. Taken together, our findings provide new evidence on the loss of fertility induced by overweight in males.

Low fertility in buffaloes with frozen-thawed semen is attributed to sperm damage that is related to formation of ice crystals during freeze/thaw process. This study was conducted to evaluate the freezability of buffalo bull semen extended with four types of extenders, including citrate egg-yolk (CEY, control), CEY with ethanolic Moringa Olifera Leaf Extract (eMOLE) as replacement of antibiotics (CEYm), eMOLE and eMOLE with conventional antibiotic (eMOLEa) with or without Coenzyme Q10 as antioxidant. Semen was subjected to calculate progressive motility, livability, abnormality, acrosome status and integrity of sperm membrane in semen after extending, equilibrating and thawing processes. Four sexually mature buffalo bulls (4-5 y and 400-450 kg LBW) were used for semen collection, and semen was collected twice weekly for 4 successive weeks. Semen ejaculates of ≥70% as mass motility percentage were pooled and extended at 37°C by the four types of extenders without or with 30 µM Co-Q10). Results cleared that progressive motility, livability, abnormality, acrosomal damage and plasma membrane integrity percentages improved (P<0.05) in buffalo semen after dilution, equilibration and thawing, while activity of AST, ALT and LDH in seminal plasma of post-thawed semen reduced (P<0.05) by extension of semen with eMOLE as a new extender. All sperm parameters and enzyme activity were maintained by replacing antibiotics in CEY extender with 4 ml eMOLE. All sperm parameters were improved (P<0.05) and enzyme activity was maintained by adding 30 µM of coQ10 as compared to free extenders. Fertility rate of buffalo cows inseminated with semen cryopreserved in extender (eMOLE+coQ10) was the highest (91.6%), followed by eMOLE without coQ10 (83.3%) and the lowest for control CEY (66.6%). In conclusion, eMOLE as a new extender, is promising for the extension and preservation of buffalo semen. Also, adding 4 ml of eMOLE could be a good substitute for the antibiotic component of CEY as a conventional semen extender. Moreover, supplementing semen extenders with antioxidant addition (30 µM CoQ10) is important to improve function of spermatozoa and sperm fertility of cryopreserved semen of buffaloes.

Fumonisins are mycotoxins produced primarily by the fungi Fusarium verticillioides and F. proliferatum, which colonize maize in tropical, subtropical, and temperate climates worldwide. Fumonisin exposure is the highest in rural populations that consume large amounts of maize and maize products. Among them, infants and young children are the most vulnerable when they are weaned onto maize‐based foods. Therefore, it is critical to understand the impact of fumonisin on children's health and growth. This review describes the evidence linking fumonisin exposure to child growth impairment. First, toxicological studies that attempt to elucidate the mechanism by which fumonisin may impair growth are discussed. Next, a description is given of candidate biomarkers for accurately capturing fumonisin exposure in humans. Potential human health effects of fumonisin exposure beyond growth impairment, including esophageal cancer and neural tube defects (NTDs), are briefly described. We review epidemiological studies that collectively show an increasing weight of evidence linking fumonisin exposure to growth impairment, particularly in world regions where young children consume maize‐based weaning foods. Additionally, the evidence linking fumonisin exposure to growth impairment in plants and a variety of animal species are reviewed. Finally, we describe interventions to reduce fumonisin in parts of the world where dietary fumonisin exposure is high and is likely to predispose populations to increased health risk.
