Epileptic seizures are seen as a result of changing excitability balance depending on the deterioration in synaptic plasticity in the brain. Neuroplastin, and its related molecules which are known to play a role in synaptic plasticity, neurotransmitter activities that provide balance of excitability and, different neurological diseases, have not been studied before in epilepsy.In this study, a total of 34 Sprague–Dawley male and female rats, 2 months old, weighing 250–300 g were used. The epilepsy model in rats was made via pentylenetetrazole (PTZ). After the completion of the experimental procedure, the brain tissue of the rats were taken and the histopathological changes in the hippocampus and cortex parts and the brain stem were investigated, as well as the immunoreactivity of the proteins related to the immunohistochemical methods.As a result of the histopathological evaluation, it was determined that neuron degeneration and the number of dilated blood vessels in the hippocampus, frontal cortex, and brain stem were higher in the PTZ status epilepticus (SE) groups than in the control groups. It was observed that neuroplastin and related proteins TNF receptor‐associated factor 6 (TRAF6), Gamma amino butyric acid type A receptors [(GABA(A)], and plasma membrane Ca2+ ATPase (PMCA) protein immunoreactivity levels increased especially in the male hippocampus, and only AMPA receptor subunit type 1 (GluA1) immunoreactivity decreased, unlike other proteins.We believe this may be caused by a problem in the mechanisms regulating the interaction of neuroplastin and GluA1 and may cause problems in synaptic plasticity in the experimental epilepsy model. It may be useful to elucidate this mechanism and target GluA1 when determining treatment strategies.

ackground: Phenobarbital is one of antiepileptic drugs. Diclofenac is one of NSAID. Objective: The purpose of carrying out this research was to determine the effect of both phenobarbital and diclofenac on the nervous convulsions resulting from the treatment of chicks with 4-aminopyridine in addition to determining the type of interaction between these two drugs. Methods: 4-aminopyridine was utilized to induce convulsion in chicks then we used Phenobarbital with diclofenac for prevent the occurrence of convulsion. Results: The chicks on which the experiments were conducted showed that the median lethal dose for 4-aminopyridine was 62.59 mg/kg i.p, while the median effective dose of 4-aminopyridine was 32.63 mg/kg. The median of anticonvulsant dose of phenobarbital in preventing nerve convulsions was 9.47 mg/kg i.m. These experiments also showed that the median of effective dose of diclofenac to prevent seizures was 7.48 mg/kg in chicks. Isobolographic analysis proved that the drug interaction between phenobarbital and diclofenac against induced nerve convulsions is synergistic interaction at 0.5:0.5 ratio this result recorded for first time in chicks, so the value of Y is lesser than one, Where the value of Y is calculated through a special equation. If the point falls on the line connecting the median doses of the two drugs, this means that the interaction is additive, and if it falls above the line, this indicates that the interaction is antagonistic, but if it falls below the line, it is synergistic interference. conclusion: our results indicate that phenobarbital and diclofenac have an effective role when used in therapy convulsion and it has synergistic interaction which is characterized by more effective in chicks in addition diclofenac has an essential side in convulsion therapy.

The present study evaluated the antiseizure and neuroprotective effects of sodium houttuyfonate (SH), a derivative of Houttuynia cordata Thunb. (H. cordata), in a kainic acid (KA)- induced seizure rat model and its underlying mechanism. Sprague Dawley rats were administered normal saline, SH (50 or 100 mg/kg), or carbamazepine (300 mg/kg) by oral gavage for seven consecutive days before the intraperitoneal administration of KA (15 mg/kg). SH showed antiseizure effects at a dose of 100 mg/kg; it prolonged seizure latency and decreased seizure scores. SH also significantly decreased neuronal loss in the hippocampi of KA-treated rats, which was associated with the prevention of glutamate level increase, the upregulation of glutamate reuptake-associated proteins (excitatory amino acid transporters 1–3), glutamate metabolism enzyme glutamine synthetase, the downregulation of the glutamate synthesis enzyme glutaminase, and significant alterations in the expression of AMPA (α-amino-3-hydroxy-5-methyl-4-isoxazole-propionic acid receptor) and NMDA (N-methyl-D-aspartic acid receptor) receptor subunits in the hippocampus. Furthermore, the effects of SH were similar to those of the antiseizure drug carbamazepine. Therefore, the results of the present study suggest that SH has antiseizure effects on KA-induced seizures, possibly through the prevention of glutamatergic alterations. Our findings suggest that SH is a potential alternative treatment that may prevent seizures by preserving the normal glutamatergic system.
