Effects of Dexmedetomidine on Immune Cells: A Narrative Review

Abstract: As we all know, dexmedetomidine (DEX), as a highly selective α2 adrenergic receptor agonist, exerts sedative, anti-anxiety and hypnotic effects by inhibiting the discharge of norepinephrine neurons in locus coeruleus and GABA-related hypnotic pathways. However, the role of DEX in anti-inflammatory and immune regulation has gradually attracted the attention of researchers in recent years. The α2 adrenergic receptor is one of the members of the adrenergic receptor family, which is widely present in a variety of … Show more

“…Nevertheless, ketamine has many other effects that may be beneficial, especially in neurosurgery, such as neuroprotection (by blockade of the N‐methyl‐D‐aspartate [NMDA] receptor), 17 antiinflammatory effects, 18,19 decreased requirement for inhalant anesthetic, 4 and decrease in postoperative shivering 20 . Dexmedetomidine also has been reported to have neuroprotective 21 and antiinflammatory 22 effects.…”

“…16 Nevertheless, ketamine has many other effects that may be beneficial, especially in neurosurgery, such as neuroprotection (by blockade of the N-methyl-Daspartate [NMDA] receptor), 17 antiinflammatory effects, 18,19 decreased requirement for inhalant anesthetic, 4 and decrease in postoperative shivering. 20 Dexmedetomidine also has been reported to have neuroprotective 21 and antiinflammatory 22 effects. Also, recent studies using experimental rodents suggested that opioids might worsen functional outcome after spinal cord injury by exacerbating the inflammation and tissue damage that follow contusive lesions.…”

“…The induction of proinflammatory cytokine and chemokines (interleukin-2) is detrimental among primary liver cancer patients who have undergone hepatectomy ( Sachdeva et al, 2015 ; Sachdeva and Arora, 2020 ; Cai et al, 2022 ; Chen et al, 2022 ; Lu et al, 2022 ). From the observations above, we could conclude that Dexmedetomidine could be a potential agent against the progression of primary liver cancer.…”

“…Dex protects cells greatly against several stresses by activating the anti-apoptotic signal pathway and considerably regulating systemic inflammatory response syndrome by decreasing the release of pro-inflammatory cytokine. In addition, Dex has been discovered to perform immunomodulatory effects by inhibiting activated immune cells such as T cells and macrophages ( Chen, et al, 2022 ). Dexmedetomidine pretreatment exerts protective efects on rat pancreas irradiated abdominally with ionizing radiation ( Mercantepe, et al, 2023 ).…”
