This study was carried out to investigate the changes in semen characteristics, hormonal profile and testicular morphometry of West African Dwarf (WAD) goat bucks treated with danazol. For this purpose, eighteen matured WAD bucks average of about two years were randomly divided into groups A, B and C comprising of six bucks per group. Group A was the control while B and C bucks were given danazol at a dose rate of 20mg/kg body weight orally daily for four weeks. The group B bucks’ testes were harvested thereafter for gross and histo-morphometric studies while those of group C were left intact but danazol treatment was withdrawn for four more weeks. The bucks’ semen samples (collected by electro-ejaculation) and hormonal samples (taken via the jugular vein) were analyzed. The semen characteristics studied included color, volume, mass activity, motility, percentage normal live-dead ratio, morphology and concentration; while the hormones studied included testosterone, follicle stimulating hormone and luteinizing hormone. Results show that there were significant decreases in semen characteristics of group B and C bucks compared with those of group A in the first four weeks. The semen characteristics of the group C bucks were reversed to normal range (similar to those of group A bucks) two weeks after danazol treatment was withdrawn. Similar results were observed with the hormonal studies. In conclusion, danazol caused a reversed reduction in sperm cells characteristics suggesting its possible use as a contraceptive in WAD goat bucks.

The authors examined the effects of the immunosuppressive drug cyclosporine (CsA) on the male reproductive system in prepubertal rats. Twenty‐one‐day‐old rats were subcutaneously injected with either cremaphorsaline vehicle or CsA (1 and 2 mg/kg/d). The animals were treated until they were 66 days old. Cyclosporine did not affect the weights of the body or testis but decreased the weights of all sex accessory organs. Quantitative analysis of the tubules in stage VII of spermatogenesis revealed a decline in the cell counts of pachytene spermatocytes and step VII spermatids. Testicular and epididymal sperm counts and motility were decreased by 50% and fertility by 60%.Cyclosporine lowered serum testosterone despite an elevation of LH, indicating that the drug directly inhibited testosterone synthesis. Serum creatinine levels were normal in the treated animals, precluding renal failure as the cause for this impairment. Intratesticular concentrations of pregnenolone and 17‐hydroxy progesterone were significantly elevated, while those of progesterone, androstenedione, and testosterone were markedly reduced. Determination of steroidogenic enzyme activities indicated that the administration of CsA inhibited the activity of Δ5‐3B‐hydroxy steroid dehydrogenase‐Δ5‐4 isomerase (3beta‐HSD). These results clearly indicate that CsA in the doses used is harmful to the male reproductive function in prepubertal rats.

Cholesterol fed to male rats and rabbits for 120 days impaired testicular function. The production of secondary spermatocytes in rats and rabbits was reduced by 26.1% and 46.9%, respectively. There was significant reduction in spermatid cell population, seminiferous tubules, and Leydig's cell nuclear dimensions. In rats aortic endothelium showed some alterations while in rabbits full-blown atherosclerotic plaque containing a necrotic core and a proliferative fibrous cap was found. Total cholesterol, triglycerides and phospholipids of the testes were increased whereas, testicular glycogen was significantly reduced. Three- and sevenfold increases were noticed in the serum cholesterol levels of rats and rabbits. The LDL cholesterol values were increased while the HDL cholesterol/Total cholesterol ratio was significantly decreased (P less than 0.001). It is concluded that hypercholesterolemia affects the testicular function and aortic endothelium.
