Effects of co-expressing the plant CDK inhibitor ICK1 and D-type cyclin genes on plant growth, cell size and ploidy in Arabidopsis thaliana

Abstract: The cyclin-dependent kinase (CDK) plays a crucial role in regulating the cell cycle of eukaryotic organisms including plants. From previous studies, it is known that ICK1, the first plant CDK inhibitor identified in Arabidopsis plants, interacts with Arath;CycD3;1 (CycD3) and Arath;CDKA;1 (Cdc2a). Overexpression of ICK1 has major effects on cell division, plant growth, and morphology. In this study, approaches were taken to determine the effects on transgenic 35S::ICK1 Arabidopsis plants of introducing another… Show more

“…Collectively, these results show that over-expression of the ICK/KRP CDK inhibitors can affect plant growth and also morphology. It should be noted that the phenotypic effects depend not only on the expression of CDK inhibitors but also the expression of interacting regulators such as D-type cyclins (Jasinski et al, 2002b;Zhou et al, 2003b). From a cell biology point of view, it is interesting to consider the effects of ICK/KRPs on important cellular processes.…”

“…CDKB1;1 , a key regulatory gene in the G2-M phase transition, has been shown to negatively regulate the onset of endocycle4. High expression level of ICK/KRP1 may suppress the endocycle and inhibit G2-M phase transition4565. RBR1 regulates the switch from proliferation to endocycles by targeting the CCS52A1 and CSS52A2 genes33.…”

“…The interactions between ICK ⁄ KRP and D-type cyclins were also observed in twohybrid targeted screens (Wang et al, 1998;Jakoby et al, 2006). Furthermore the in planta interaction was indirectly demonstrated by the simultaneous overexpression of AtCycD3;1 and NiKIS1a from tobacco, as a wild-type leaf phenotype was restored in the double overexpressing plants (Jasinski et al, 2002b;Schnittger et al, 2003;Zhou et al, 2003a). In addition, SlKRP1 is able to import SlCDKA;1 into the nucleus (Fig.…”
