Effects of cellular iron deficiency on the formation of vascular endothelial growth factor and angiogenesis.

Abstract: BackgroundYoung women diagnosed with breast cancer are known to have a higher mortality rate from the disease than older patients. Specific risk factors leading to this poorer outcome have not been identified. In the present study, we hypothesized that iron deficiency, a common ailment in young women, contributes to the poor outcome by promoting the hypoxia inducible factor-1α (HIF-1α and vascular endothelial growth factor (VEGF) formation. This hypothesis was tested in an in vitro cell culture model system.Re… Show more

“…The down-regulation of myelination-related genes and up-regulation of those related to angiogenesis is in line with previous reports that ID leads to hypo-myelination and increased angiogenesis (Beard et al, 2003; Todorich et al, 2009; Eckard et al, 2010). Hypo-myelination is also characteristic of RLS in humans (Connor et al, 2011).…”

“…However, TfR1 expression is also regulated at the transcriptional level. TfR1 expression is up-regulated by the hypoxia-inducible factor (6), which is typically activated under hypoxic conditions (7), but can also be turned on by a number of non-hypoxic stimuli, including inflammatory signals such as nitric oxide and l/opolysaccharide in macrophages (8) and 17␤-estradiol in estrogen receptor-positive cells (9).…”

“…This limitation might affect the specificity of proposed criteria and a validation study will be required. We also have to raise false-positive conditions potentially associated with elevated serum VEGF, including iron deficiency,23 anaemia,24 hypoxia25 and various types of cancer 25. Patients with breast, gastrointestinal or prostate cancer present further increase in VEGF levels on metastatic stage 25.…”
