Effects of Candesartan on Electrical Remodeling in the Hearts of Inherited Dilated Cardiomyopathy Model Mice

Abstract: Inherited dilated cardiomyopathy (DCM) is characterized by dilatation and dysfunction of the ventricles, and often results in sudden death or heart failure (HF). Although angiotensin receptor blockers (ARBs) have been used for the treatment of HF, little is known about the effects on postulated electrical remodeling that occurs in inherited DCM. The aim of this study was to examine the effects of candesartan, one of the ARBs, on cardiac function and electrical remodeling in the hearts of inherited DCM model mi… Show more

“…Our study showed that the heart rate at 21 d in the CDs group was significantly higher than that of the saline group, indicating that CDs had some toxicity in the heart, a result similar to other groups' findings [31,32] . However, the heart rate of the PEG-CDs group was similar to that of the saline group, suggesting the PEGylation could considerably attenuate the heart toxicity of CDs.…”

“…Candesartan was administered at 1 mg/kg bw/day (9.42 mg drug/kg diet). This dose suppresses the growth of human bladder carcinoma cell xenografts in nude mice by 42 % (Kosugi et al 2009 ) and increases the survival of a mouse model of inherited dilated cardiomyopathy (Odagiri et al 2014 ). The dosages used were chosen for the likelihood that they would be both safe and effective at producing a relevant biological response over the lifespan of a treated mouse.…”

“…Reduced AT1R signaling reduces blood pressure and the incidence of stroke, diabetic kidney disease, congestive heart failure, diabetes mellitus, and atrial fibrillation [reviewed in (Strauss and Hall 2016 )]. Until recently, angiotensin II receptor antagonists (ARAs) were thought to have effects very similar to those of ACEis (Yusuf et al 2003 ; Odagiri et al 2014 ). More recent meta-analyses suggest that ACEis, but not ARAs, reduce myocardial infarction and all-cause mortality in patients with hypertension (van Vark et al 2012 ; Strauss and Hall 2016 ).…”

“…It has also been shown to be expressed in the human left ventricle [ 26 ] and would be functional in canine ventricular myocytes [ 27 ]. It was observed in mice ventricular myocytes, contributing to repolarization [ 28 ]. However, its contribution to repolarization in human or pig ventricles remains to be demonstrated.…”
