Effects of brotizolam on the sleep of chronic insomniacs.

Abstract: 1 Effects of 0.5 mg brotizolam on the sleep of chronic insomniacs were assessed electroencephalographically and subjectively over 14 days. 2 Brotizolam (0.5 mg) increased total sleep time, decreased drowsy (stage 1) sleep and increased stage 2 sleep. At this dose it also decreased slow wave and rapid eye movement sleep. On withdrawal there was evidence of insomnia in some subjects during the first night. The drug was well tolerated. 3 Further studies are indicated with lower doses of the drug.

“…However, unlike the case with many short-acting benzodiazepine hypnotics, with drawal insomnia was not found with zopi clone [5,6], This confers an important clini cal advantage on this drug. The effects of zopiclone on sleep morphology were consis tent with its short half-life and were compa rable to those of short-acting benzodiazepine hypnotics [7,8], Zopiclone decreased the du ration of stage 1, increased the duration of stage 2, and slightly, but definitely, decreased the duration of stages 3 and 4. There was no significant effect on REM sleep.…”
